| In the paper, the part of chemical components of Usnea longissima Ach. were isolated and identified, and two of them had significant tumor cells inhibition effects after 48 hours compared with blank control(P < 0.01). the dosage-dependent effect between concentration and inhibition effects was existed. A preliminary study of its anti-tumor mechanism.The powder of Usnea longissima Ach were extracted by petroleum ether(30-60), dichloromethane and ethyl acetate. Three compounds were isolated and identified, they were usnic acid,(Z)-2-acetyl-5, 5-bis(7-acetyl-4, 6-dihydroxy-3, 5-dimeth-ylbenzofuran-2-yl)-4-hydroxypenta-2, 4-dienal and ethyl 4-(7-acetyl-4, 6-dihydroxy-3, 5-dimethyl-2-oxo-2, 3-dihydrobenzofuran-3-yl)-4-(7-acetyl-4, 6-dihydroxy-3, 5-dimethylbenzofuran-2-yl)-3-oxobutanoate.The two optimized compounds were used to do further study by Affymetrix gene chip on HepG2. Differential expression, gene ontology and pathway were analyzed with a lot of deta in this part. The significant analysis of microarrays was done with fold-charge and pvalue as a standard.(Z)-2-acetyl-5, 5-bis(7-acetyl-4, 6-dihydroxy-3, 5-dimeth-ylbenzofuran-2-yl)-4-hydroxypenta-2, 4-dienal was analyzed with fold-charge greater than 2 and pvalue less than 0.01, there were 246 up-regulated expression genes in 482 significant changed genes. The six tumor related signaling pathway, cell cycle signaling pathway, endocytosis signaling pathway, pathways in cancer, prostate cancer signaling pathway, p53 signaling pathway and colorectal cancer signaling pathwaywere significant changed. 266 Biological Process, 68 Cellular Component and 43 Molecular Function were attended by significant expressed genes in gene ontology analysis. ethyl 4-(7-acetyl-4, 6-dihydroxy-3, 5-dimethyl-2-oxo-2, 3-dihydrobenzofuran-3-yl)-4-(7-acetyl-4, 6-dihydroxy-3, 5-dimethylbenzofuran-2-yl)-3-oxobutanoate was analyzed with fold-charge greater than 2 and pvalue less than 0.01, there were 8 up-regulated expression genes in 104 significant changed genes. The two tumor related signaling pathway, cell cycle signaling pathway, endocytosis signaling pathway significant changed. 109 Biological Process, 48 Cellular Component and 15 Molecular Function were attended by significant expressed genes in gene ontology analysis.Both are mainly through endocytosis and cell cycle work.To filter out a significant expression differences genes: TXNIP;PTEN;EGFR;CASP9;DDIT3;HSPA8;GPNMB;AKR1C1;ALCAM,RAD21;PRKDC;AKAP9;C14orf106;TXNIP;STAG2;ITGA6;IQGAP1, by realtime qPCR, Verify whether it can induce these genes up or down regulated, The results show, TXNIP,PTEN,CAPS9,DDIT3,AKR1C1,STAG2,IQGAP1,RAD21,C14orf106 increase, EGFR,HSPA8,GPNMB,ALCAMã€PRKDC,ITGA6,AKAP9 down. Among them TXNIP,CASP9,AKR1C1 are more obvious changes,reaching more than 500%.DDIT3,RAD21,C14orf106 are raised moderate changes, PTEN,STAG2,IQGAP1 have small changes. |
