Yinqiao Jiedu Pill is kind of cold-pungent and diaphoretic Chinese patent medicine with the effect of removing heat and toxic substance from the body, consisting of Lonicera japonica flos, Forsythiae fructus, Menthae haplocalycis herba, Schizonepetae herba, Sojae semen praeparatum, Arctii fructus(parched product), Platycodonis radix, Lophatheri herba and Glycyrrhizae radix et rhizome. It is a common Chinese drug for the treatment of wind-heat type of common cold with the symptoms of fever, headache, cough, dry mouth and sore throat. And it is widely used in the clinical application due to its stable therapeutic effect. However, only qualitative detection of microscopic identification and TLC without quantitative item presented in the quality standard of Yinqiao Jiedu Pill, which could not reflect the comprehensive quality circumstance of this preparation.In this paper, a rapid and sensitive HPLC method was established for simultaneous determination on chlorogenic acid, phillyrin, arctiin, liquiritin and glycyrrhizic acid in Yinqiao Jiedu Pill by systematic investigation of sample preparation method and chromatographic method as well as methodology validation with the study object of Yinqiao Jiedu Pill(Water-honeyed pill).Different extraction conditions including extraction solvent, extraction method, extraction time and solvent volume were investigated. Finally, the extraction condition was selected as ultrasonation with 25 m L of 70 percent of methanol for 30 min. In addition, the chromatography condition was optimized as follows: Chromatography column: Thermo C18(4.6 mm*250 mm, 5 μm); Mobile phase: acetonitrile-0.5 percent of phosphoric acid water solution; Elution method: gradient elution; Flow rate: 0.8 m L/min; Detection wavelength: 230 nm; Column Temperature: 25℃. Results of methodology validation suggest that the detection components of chlorogenic acid, phillyrin, arctiin, liquiritin and glycyrrhizic acid were in good linear correlation in the amount range of 0.1628-1.9536μg, 0.0108-0.1301μg, 0.2024-2.4288μg, 0.0502-0.6024μg and 0.1582-1.8984μg, respectively, and the correlation coefficient value was greater than 0.9997. The average recovery rates of these five components were 99.42%(RSD = 0.53%), 100.96%(RSD = 1.05%), 100.82%(RSD = 0.71%), 100.64%(RSD = 0.78%), 99.02%(RSD = 1.72%).This method was simple, rapid and sensitive with good accuracy and repeatability, which could be used in the determination of chlorogenic acid, phillyrin, arctiin, liquiritin and glycyrrhizic acid in Yinqiao Jiedu Pill(Water-honeyed pill), and provide some theoretical evidence for the quality evaluation for Yinqiao Detoxication Pill and its relational preparations.
