Objectives Chronic periodontitis is a chronic infectious disease which caused by plaque biofilm in periodontal supporting tissue(gums, alveolar bone and periodontal membrane,etc.), its development is closely related to systemic diseases, such as diseases of the cardiovascular system. Chronic inflammation is recognized as a major driving force in atherogenesis. And CP may be closely related with the AS development and is one of the independent risk factors for the AS development of disease. High sensitive C-reactive protein(hs-CRP) is an important biomarker of inflammation and the strong predictive factor of cardiovascular events. It played an important role in the development of CP and AS. This study is regarded to set up the experimental animal models of CP and AS, to observe the effect of Gegen Qinlian decoction on high sensitive C-reactive protein in periodontitis and high sensitive C-reactive protein in periodontitis and atherosclerosis compound model in mice, to investigate the relationship between serum levels of hs-CRP with periodontitis and atherosclerosis, to observe the treatment effect of Gegen Qinlian decoction on periodontitis and atherosclerosis compound model in mice, therefore, we would provide experimental basis for clinical application of Gegen Qinlian decoction in the treatment of patients who suffered from periodontitis and atherosclerosis.Methods 46 male SD Wistar rats were randomly divided into two groups: group A(normal control group of 8, ordinary water feeding), group B(periodontitis and atherosclerosis compound model group of 38). The method of establishing an animal model of chronic periodontitis accompany with atherosclerosis in Wistar rats was to ligated the left maxillary second molars by steel wire ligation and fed with high glucose and lysophosphatidylcholine diet for 12 weeks. At the 12 th weekend, rats of normal group A and model group B were randomly executed only 3 in order to verify the model’s establishment. After the success of the modeling, group B were randomly divided into 5groups given drugs for 4 weeks: saline group(group B1), atorvastatin group(group B2),metronidazole group(group B3), atorvastatin + metronidazole group(group B4) and Gegen Qinlian decoction group(group B5). After 4 weeks, we examined the SBI、PD of the experimental animals, and then, all the rats were sacrificed and taken samples. Serum hs-CRP levels were assayed by ELISA. Attachment loss level and vascular change were examined by pathology.Results 1 Clinical examination results revealed: during the experiment, the rats in the model group were significantly diet reduction and weight loss, the periodontal ground showed acute inflammatory response and their periodontal showed red and swollen obviously, SBI and PD gradually get worse. After treatment, different group’s animals periodontal inflammation improved better, and group B4, B5 were significantly(P<0.05).2 ELISA results show: The levels of serum hs-CRP in group B1-5 were significantly igher than that of group A(P<0.01). After medical intervention, serum hs-CRP levels of group B2 and B3 were significantly lower than that of group B1(P<0.05) and higher than that of group B4 and B5(P<0.05), but there was no statistical difference between group B2 and group B3(P>0.05). Serum hs-CRP levels of group B4, B5 were significantly lower than that of group B1-3(P<0.01), but there was no statistical difference between group B4 and group B5(P>0.05). 3 Histopathological observation of periodontal tissue revealed: the inflammation of periodontal tissues in group B2-5 were improved better than that of group B1, and group B4, B5 were significantly. Attachment loss levels in group B4 and B5 were lower than that in other groups, but there was no statistical difference between group B4 and group B5(P>0.05). 4 Histopathological observation of arteries revealed: sharp thickness of artery wall, formation of foam cell, disorganized muscle fiber in model group,Compared with group B2 and B3, haemorheological disorder and atherosclerotic changes were much improved in group B4 and B5 after medical intervention, there was no foam cell in group B4 and B5, the thickness of artery wall in group B4 and B5 much even and muscle fiber were arranged much regular.Conclusions 1. Simple periodontal ligation of the maxillary second molars and high glucose and lysophosphatidylcholine diet can establish chronic periodontitis and atherosclerosis compound model in mice in 12 weeks, method is simple and stable. 2. The high level of hs-CRP played an important role in promoting the development of CP and AS.3. Gegen Qinlian decoction can significantly improve the degree of pathological injury in chronic periodontitis and atherosclerosis compound model in mice. No significant difference in the treatment effect with atorvastatin and metronidazole group. 4. The Gegen Qinlian decoction could inhibit the development of chronic periodontitis and atherosclerosis through reducing the expression of serum hs-CRP.
