Objective : Human beta-defensin(h BD) is an important substance to resist bacterial infection. It not only expressed in respiratory tract, reproductive epithelium, adult heart, skeletal muscle, fetal thymus, esophagus, trachea and placental membrane, but also in the human oral cavity. Recently, the research on human beta defensin in the human oral cavity have made great progress. Through these studies, we can explore new path for the treatment of oral diseases.Methods:After the genes of h BD-3 and GFP transferred into the cells, the polymerase chain reaction assay and western blot assay have been utilized to detect the secreted h BD-3from cells. After those assays confirmed, we used the green fluorescence microscope counting cells. Then the capacity to generate the new bone tissue has been checked.Results :The efficiency of transfection by using lentivirus which contain h BD-3 and GFP genes effect on the h BMCs and h PDLCs is 44.44% and 28.98%. The efficiency of transfection by using lentivirus which contains GFP gene effect on the h BMCs and h PDLCs is79.94% and 31.71%. All the transfection efficiency mentioned above are considerable.Both of the cells which have been transferred by lentiviralscan generate new bone still.Conclusions: By using the method of lentivirus transfection, the h PDLCs and h BMCs can make the h BD-3 gene over expression. The transfection efficiencies are similar. Above all,h PDLCs and h BMCs can be the ideal candidate for periodontal tissue regeneration. The cells are still has the capacity to generate the new bone.
