Plumeria rubra Linn. cv. Acutifolia(Apocynaceae) has the effects of “clearing-heat” and “inducing-diuresis”. Plumeria rubra Linn. cv. Acutifolia has been widely used for its high ornamental value and the flower of Plumeria rubra Linn. cv. Acutifolia(FPR) are used as tea, food and cosmetics, etc.In recent years, a larger amount of Plumeria Flos are consumed increasingly because of the rise of the herbal tea industry. However, there are only characteristics description of current quality standard in China. Therefore, it is necessary to investigate more scientific and more comprehensive quality control method to solve this problem. Because of current quality standard is lack of the specific identification and effective ingredient content determination. Therefore, this thesis was carried out on Plumeria rubra Linn. cv. acutifolia and similar species using microscopic identification, TLC identification, phytochemical identification and DNA barcoding. The result is as follow:① Microscopic identification: A large amount of nonglandular hair with exclusive characteristics was observed.② Thin-layer chromatography(TLC): The quality control method for essential oil, triterpene and flavonoids were respectively built. Geraniol and nerolidol were identified in the essential oil; Lupine alcohol was identified in triterpene; Quercetin and naphthol were identified in flavonoids; β-sitosterol was identified in sterols. In conclusion, TLC can distinguish Plumeria rubra Linn. cv. acutifolia and Plumeria rubra Linn well.③ Gas chromatography(GC) was used to establish the content determination method for volatile component including linalool, geraniol and nerolidol. The application of high performance liquid chromatography(HPLC) was used to establish the content determination method for lupeol. The results showed that lupine alcohol content was as high as 0.3%. The quercetin and kaempferol were also determined that the content of kaempferol was as high as 0.4%.④DNA barcoding approach was used to identify Plumeria Flos from gene level to establish the DNA barcode. Four candidate sequences ITS2, psbA- trnH, matK and rbcL were examined. Selected psbA- trnH to DNA barcoding after experiment. Result provided that the frangipani intrinsic molecular information. Besides, the results showed that the morphology has intrinsic relationship with its genetic information. This molecular biological methods provided an effective method for identification research of Plumeria rubra Linn. cv. acutifolia, and it is proved to be an effective complement method for traditional pharmacognostic identification.This thesis used microscopic identification, phytochemical analysis methods and molecular biological technology to enrich the quality standard and solved the problem of authenticity identification. In addition, this study provides a new thought for comprehensive subject results to improve the quality standard.
