Agarwood, as the traditional Chinese medicine and widely distributed Southeast Asian countries, is a resinous wood from the tree of Aquilaria sinensis(Lour.) Gilg(Thymelaeaceae). It has a long history of application in China.This study took the extract of agarwood as the research object, which was extracted by different solvent, got the different polar extracts(EtOAc extract, n-BuOH extract and aqueous extract) and then obtained the bioactive extract through the investigation of the antimicrobial activity of extracts. Finally, we found the antimicrobial activity of agarwood concentrated in the Ethyl acetate part.25 compounds were isolated from the antimicrobial part and 22 of them were determined. On the basis of spectroscopic analysis including UV, 1D, 2D-NMR and MS,their structures were identified as Baimuxinal(1), Baimuxinic acid(2),(+)-Selina-4,11-dien-14-al(3), 4-(4-hydroxy-3- methoxyphenyl)-butanone(4), Vetaspira-2(11),6-dien-14-al(5), 3-Isopropenyl-4a,5-dimethyl-octahydro-naphthalene-2,8a-diol(6), 4-hydroxyphenyl propionic acid(7), Baimuxinol(8), 6-Methoxy,7-Hydroxy-2-[2-(4-methoxyphenyl)ethyl]-4H-chromen-4-one(9), 6,8-Dihydroxy-2-(2-phenylethyl)-4H-chromen-4-one(10), Vanillin(11), 2-[2-(4-methoxyphenyl)ethyl]-4H-chromen-4-one(12), β-Agarofuran(13),(-)-10-Epi-γ-eudesmol(14), 6-Hydroxy-2-[2-(4-methoxyphenyl)ethyl]-4Hchromen-4-one(15), 5,8-Dihydroxy-2-(2-phenylethyl)-4H-chromen-4-one(AH7)(16), 5-Dihydroxy-8-methoxy-2-(2-phenylethyl)-4H-chromen-4-one(17),6-Methoxy-2-[2-(4-ethoxyphenyl)ethyl]-4H-chromen-4-one(18), 8,8a-Dimethyl-1,7,8,8a-tetrahydro-naphth alene-2,6-dione(19),nor-Keto-agarofuran(20), octadecane(22), nonacosane(22). Compound 6 and 17 are a new compound.Though the antibacterial activity test of the compounds isolated, the results indicated that 9, 15, 16 and 18 showed inhibitory activities against Staphylococcus aureus in varyingdegrees; compound 6, 9, 15, 16 and 17 have better inhibitory activities against Anti-methicillin-resistant Staphylococcus aureus than others.This study was conducted to evaluate the effect of three different approaches(HAc,HAc+ Fusarium SP A2 and HAc+ Botrosphaeria rhodina A13 for artificial inducing the formation of twelve batches of agarwood, with thirty-six samples using antimicrobial activity. The antimicrobial activity was determined by a two-fold serial dilution method and minimum inhibitory concentration(MIC) against a panel of microorganisms. The results show that the HAc + A2 and HAc + A13 approaches for artificial inducing the formation of agarwood at 12 mouths which were batter. This research results were consistent with the experimental results by our previous chemical methods. Finally, we have reached the conclusion that artificial agarwood obtained by comprehensive stimulated method(formic acid plus fungal inoculation) and longer induced time have better antimicrobial activity.This paper is a beneficial exploration to the first use of antimicrobial activity to evaluate artificial agarwood obtained by different producing methods and different culture time.
