As rapid, effective, nondestructive optical techniques, Raman and SERS spectroscopy have been successfully employed in the field of biomedicine in recent years. The secreted proteins have been a hot research topic in biomedical field with more realization about the function of secreted proteins of cancer cells. Generally, the conventional biological methods for analysis of secreted proteins require complex operation steps and organic solvents often contain some toxicants. This may be time consuming and troublesome. Therefore, it is necessary to find a new method for rapid and effective analysis of secreted proteins. In this study, Raman and SERS spectroscopy were applied to detect the secreted proteins of hepatoma cells and normal liver cells, and the human plasma proteins. In addition, we used PCA and cluster analysis to discriminate the spectra between different groups. The main research results are follows:1. Raman spectroscopy and SERS spectroscopy were used to detect the secreted proteins of human hepatoma cell line HepG2 and normal liver cell line LO2, PCA-LDA method were applied to analyze the data of regular Raman spectra with 83.3% sensitivity and 100% specificity and SERS spectra with sensitivity 100% and specificity 100%, the results show that the regular Raman spectroscopy and SERS spectroscopy both have potential for detection of secreted proteins.2. SERS spectroscopy was applied to analyze the difference of component and conformation in secreted proteins from HepG2 cells after the treatment of doxorubicin in different concentration. The analysis of PCA-LDA can distinguish the spectral data well with high sensitivity and specificity.3. We focused on plasma proteins from 20 live cancer patients and 20 healthy subjects and compared the Raman spectra and SERS spectra between two groups. The results of PCA showed that the Raman spectra of plasma proteins from liver cancer patients and healthy volunteer can be discriminated with high sensitivity and specificity.
