Objective To Analysis affects of Yiaikang Capsule on asymptomatic HIV infection the Th9 cells and expression of transcription factor PU.1, to explore the mechanism of Yiaikang capsule in the treatment of HIV.Methods This experiment chose 30 patients who were confirmed by HIV antibody test positive, during March to May of 2014 from AIDS Research Center, the First Affiliated Hospital of Henan University of Traditional Chinese Medicine.The HIV patients were divided into Traditional Chinese medicine Yiaikang Capsule, Collect samples.And at the same time,30 healthy people selected from medical examination center as healthy controlgroup. Yiaikang Capsule group was treated with Yiaikang capsule treatment For patients with HIV,3 times a day, each time 5, Half a year after treatment, Collect samples.again, Use Percentages of lymphocyte subsets (CD3+T cells, CD3+CD8+T cells, CD3+CD4+T cells, CD3-CD16+CD56+ cells, CD3-CD19+ cells),Th9 (CD4+IL-9+), Th1 (CD4+IFN-γ+) andTh2 (CD4+IL4+) cells were investigated by Flow cytometry (FCM) The expression of PU.1 mRNA, T-bet mRNA and GATA3 mRNA levels were detected by reverse transcription-polymerase chain reaction (RT-PCR). To analysis on the Yiaikang intervention, Th9 cells in the pathogenesis of HIV/AIDS patients and the relationship between disease progression.Results 1. Analysis of lymphocyte subsets:HIV infection before and after treatment, Percentage of CD3+CD4+T cells of HIV patients were significantly lower than the healthy control group (P<0.05), percentage of CD3+CD8+T cells of HIV patients were significantly higher than the healthy control group (P<0.05).There was no significantly different of CD3+T cells, B (CD3-CD19+) (10.52±8.38)cells and NK (CD3-CD16+CD56+) (18.53±13.81)cells ratios between the four groups (P>0.05)2.Analysis of Thl/Th2/Th9 cells and the corresponding transcription factor:A group aftertreatment of Th1 cells (7.06±11) percentage of CD4+T cells was higher than that before treatment (5.45± 7.20) had statistical significance (P<0.05).The Expression of transcriptionfactor T-bet mRNA (10.83±8.78) higher than that before therapy (8.34±6.61), there is statistical significance (P<0.05).Higher than that in healthy control group (P<0.05).After treatment of Th2 cells (0.56±0.95) percentage of CD4+T cells compared with before treatment had no statistical significance (P>0.05).Expression of Th2 cell transcription factors GAT A3 and mRNA (1.98±0.86) showed no statistical significance with before treatment (P>0.05).After treatment of Th9 cells (1.24±1.50) percentage of CD4+T cells is lower than before treatment (2.07±2), the difference was statistically significant,The Expression of transcription factor T-bet mRNA (2.39±1.51) lower than that before therapy (3.01±1.36), there is statistical significance (P<0.05)Conclusion 1. The balance between Thl and Th2 was broken in HIV/AIDS, Thl cells down, Th2 cells uper, and their transcription factor T-bet and GATA3 had changed with the progression of disease.2. Overexpression of PU.1 mRNA, T-bet mRNA and GATA3 mRNA revealed that Th9, Th1 and Th2 cells involved in the pathogenesis of HIV/AIDS.3. Yiaikang can reduce Th9 cells and expression of transcription factor in the peripheral blood of HIV/AIDS patients, Improve HIV/AIDS patient’s immune function.
