Foreword Studies have shown that,Yiaikang capsuleintervention treatment can improve the syndrome of TCM performance in patients with HIV /AIDS patients, stable or slow down the decline in C D4+T lymphocytes, stable or lower viral load, so as to improve their life quality and prolong the survival period of AIDS, delaying the development process. Human immunodeficiency virus infection is immune to damage process of a slow progress.CD4+T lymphocytes are the main target cells for HIV infection, which play a key role in the progression and treatment of HIV infection in the pathogenesis. Regulatory T cells(Treg), T helper 17 cells(Thl7) as a recently discovered two new species of T lymphocyte subsets, the differentiation, development mechanism is different from Thl, Th2, and autoimmune diseases and infections were closely related to the development. Research has shown that, in the pathogenic SIV infection animal model and HIV infection of Treg/Thl7 imbalance, there are obvious whether peripheral blood or intestinalmucosa.Objective 1.Analysis content of HIV or AIDS infected persons(HIV/AIDS) before and after administration of Yiaikang capsule in peripheral blood mononuclear cells(PBMC) regulatory T cells(Treg) and T helper 17 cells(Thl7), preliminary study the effect of Yiaikangin patients with HIV/AIDS in Th17/Treg cells.2. Analysis the amount of expression of HIV/AIDS patients before and after administration of Yiaikang capsule in peripheral blood of transcription factor RORγt m RNA, Fox P3 m RN A, preliminary study the effect of Yiaikang on HIV/AIDS in patients with Th17/Treg cell transcriptionfactor.MethodsScreening at the beginning of 2014 the First Affiliated Hospital of Henan University of Traditional C hinese Medicine during the period of AIDS research center by HIV antibody confirmation test is positive and taken alone in 47 cases of Yiaikang. six months before and after treatment of patients,according to the HIV/AIDS patients were divided into A, B two groups, 20 cases of healthy people over the same period were se lected from medical examination center as healthy control group, using flow cytometry(FCM) the detection of lymphocyte subsets and CD4+T cells, Th17(C D4+ IL-17+)/ Treg(CD4+CD25+Foxp3+) cell percentage. RT-PCR detection of peripheral blood mononuclear cells(PBMC) transfer expression transcription factor RO R gamma t m RNA, Fox P3 m RN A.Results 1.Analysis of lymphocyte subsets: group A(before treatment), group B(medication) CD4+T cells in patients with(339.36 ± 165.49, 348.47 ± 165.79) were significantly lower than that in healthy control group( 832.05 ± 106.25)( P<0.01). B group(medication) CD4+T cells in patients with(348.47 ± 165.79) higher than that of A group(before treatment) patients(339.36 ± 165.49)(P<0.01).2.Analysis of Th17, Treg cells and their corresponding transcription factors: A group(before treatment), B group(medication) in patients with Th17 cell percentage of CD4+T(1.90±3.55, 2.13 ±4.11) and healthy controls(2.68 ±1.25) contrast, have significant difference(P<0.05); A group(before taking the medicine the percentage of CD4+T cells) in patients with Th17( 1.90± 3.55) lower than that of B group( medication) patients(2.13 ± 4.11)(P<0.05); A group(before treatment),B group(medication) in patients with Treg cell percentage of CD4 + T( 3.59± 3.19, 3.37 ± 2.96) were significantly higher than that inhealthy control group(0.36± 0.27)(P<0.01); group A(before treatment) of patients with Treg cell percentage of CD4+T(3.59± 3.19) higher than that of B group(medication) patients(3.37± 2.96)(P<0.05).Group A(before treatment), group B(medication)relative expression of Th17 cell transcription factor RO R gamma t m RNA(3.18±1.15, 3.35±1.15) were lower than that of healthy controls(7.24±2.25)(P<0.05); group A(before treatment)(3.18±1.15) was lower than that of group B(3.35±1.15) after medication); group A(before treatment), group B( medication) relative expression of Treg cell transcription factors Fox P3 and m RN A(3.07 ± 1.98, 2.72 ± 1.89) were higher than those in healthy control group(P<0.05); group A( before treatment)(3.07± 1.98) higher than that of group B(after taking the medicine)(2.72 ± 1.89)(P<0.05);3.The correlation between Th17 cells and ROR gamma t m RNA : Th17 cells associated with RORgamma t m RNA positive(R =0.574, P<0.05).4.The correlation between Treg cells and Fox P3 m RNA cells: Treg associated with Fox P3 m RNApositive(R =0.532, P<0.05).Conclusion1. The percentage of Th17 cells in HIV/AIDS patients was lower than normal persons,The percentage of Treg cells in patients with HIV/AIDS is higher than normal persons, this lead to the imbalance of Th17/Treg cells. Yiaikang capsule has the role of regulating this imbalance.2. The content of RORγt m RNA of HIV/AIDS patients is lower than normal persons, the content of Fox P3 m RNA of HIV/AIDS patients is higher than normal persons, this Lead to imbalance.
