Objective: Chinese herbal medicine is the treasure of our Chinese nation, has thousands of years under the guidance of theory of traditional Chinese medicine for the treatment of human diseases, and has the outstanding contributions. About more than 100 kinds of podocarpaceae species in the genus podocarpus,It is widely distributed in tropical, subtropical and the south temperate zone, It is mainly distributed in the southern provinces in our country.Podocarp Fruit is the seed and receptacle of the Podocarpus macrophyllus.In the book of Chih-wu ming-shih t’u kao,It has been described as such, "Podocarp Fruit in Yunnan province, with the thumb size, the seeds of green, red receptacle, the shape is very good-looking, receptacle tastes fresh and tender sweet, as the saying goes, eat this benefit a lot". Now the domestic and foreign scholars have done a lot of research about it,it has been found that it has obvious antioxidant activity. These studies illustrate Podocarp Fruit may develop into a new class of reducing blood fat, liver and antioxidant drugs.In this experiment, This study based on the theory of pharmacology of Chinese medicine, under the guidance of modern medical theory, through the simple and reliable model of hyperlipemia mice to observe Podocarp Fruit different concentrations of ethanol extracted parts influence on blood lipid in mice hyperlipidemia model, study and determine podocarpus real fall hematic fat, anti-oxidation and protect liver to protect liver effective parts, by means of molecular biology, to further study the mechanism, thus for the cholesterol-lowering podocarpus provide theoretical basis for the clinical reasonable application., studies and determine Podocarpus solid fat, antioxidant and hepatoprotective effective part, and discuss related mechanisms in order for the clinical application of Podocarpus provide the basis for further solid fat.Methods: 60 male SPF Kunming mice, were randomly divided into 6 groups: normal group,the positive group, model group,50% Ethanol Extract from Podocarp Fruit group, 75% Ethanol Extract from Podocarp Fruit group, 95% Ethanol Extract from Podocarp Fruit,The alcoholic extract of group Podocarp Fruit, each group have 10 mice. The normal group was fed with normal diet, and other groups were fed with High-fat Food(HFD) in the first 4 weeks. After 4 w, the normal group and model group mice were fed with 0.4 ml of distilled water only. While the positive group mice were fed with Simvastatin with a dose of 6.7mg·kg-1·d-1,. After administration, the determination of serum total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL-C), low-density lipoprotein cholesterol(LDL-C), alanine transaminase(GPT / ALT), aspartate transaminase(GOT / AST), malondialdehyde(MDA), superoxide dismutase(SOD) vitality, and liver MDA, SOD, organ index calculation, atherogenic index(AI), and observed in liver biopsy. While weighing and calculating the liver, spleen, kidney organ coefficient, and part of the liver samples were taken for the production of pathology and genetic analysis of PCR.Results: After the high-fat diet of 4 weeks,compared with the normal group,the level of TC, TG and LDL-C in model group has a very significant increase(P <0.01), while the HDL-C was significantly decreased(P <0.05), indicating the model of hyperlipidemia successfully constructed.1, the effect on hyperlipemia mice blood lipids related indicators: 50%, 75% and 95% alcohol extraction and alcohol leaching to decrease TC, TG, LDL- C all have different degrees of influence, and 95% alcohol extraction part works best, achieve significant difference(P < 0.05), close to the three indicators in positive control group content in the serum in mice. 2, the impact on liver function in hyperlipidemic mice:the 95% Ethanol Extract from Podocarp Fruit, significant liver average coefficient of 95% ethanol extract sites and alcohol soaked matter HLP group of mice, ALT and AST enzymes reduce water(P <0.05 or P <0.01), and 95% ethanol extract of Podocarpus real part in improving the effect is most obvious(P <0.01).3, the effect on hyperlipemia mice antioxidant capacity: compared with model group, four group, only two indicators of the 95% alcohol extraction group have significant difference(P < 0.05), and close to the value of the positive group. Compared with model group, Podocarp fruit of different concentration alcohol extraction and alcohol leaching content in mice liver SOD enzyme activity were significantly higher(P < 0.01), only 95% alcohol extraction group, the content of MDA significantly is lower(P < 0.01). 4, the molecular mechanism: Compared with the modelgroup,the Apo A-I m RNA of positive group and 95% alcohol extraction relative expression levels were significantly up-regulated(P <0.01). Meanwhile, LDL-R m RNA relative expression levels were significantly up-regulated(P <0.05). Liver biopsy HLP each group of mice further validate these results. We concluded that 95% alcohol extraction was the liver effective parts.Conclusion: 1.The high-fat diet which used in this experiments can significantly increase the mouse,s weight and content of total cholesterol in serum,and can also cause severe fat pathological change.This indicates that the model is feasible and agree with the purpose of this paper. 2.95% Ethanol Extract from Podocarp Fruit can significantly reduce weight of the HLP mice and the biochemical indexes in the serum. Therefore, 95% ethanol extraction from Podocarp Fruit is the effective parts of Podocarp Fruit in reducing plasma lipid. 3.95% Ethanol Extract from Podocarp Fruit is the effective parts of Podocarp Fruit in antioxidant and liver protection. 4.The mechanism of 95% ethanol extraction from Podocarp Fruit may be by increasing HDL- C through increasing the expression of apolipoprotein,s key Apo A-I m RNA. At the same time the 95% ethanol extraction from Podocarp Fruit will reduce LDL- C by increasing the expression of LDL-R m RNA in the HLP mice,sliver. Therefore,the plasma lipid in HLP mice can be reduced through those dual function. 5. The experiment confirmed that n95% ethanol extraction from Podocarp Fruit is the effective part of Podocarp Fruit, which provided theoretical reference for its future drug production process.However, this part is a collection of a group of compounds, it also requires a detailed analysis of its associated components. Further use of extraction and separation technology is recommended, as well as by means of analysis of LC-MS / MS, etc. to gradually narrow the pharmacodynamic material basis of Podocarp Fruit.
