With the increasing development of the antioxidants and human safety awareness, synthetic antioxidants brought increasingly prominent side effects. A natural antioxidant activity of plant extracts become a hotspot of modern scientific research. Previous studies have found a good antioxidant in a variant of this issue chrysanthemum- Shennong Xiangju which as the research object. Firstly,combining headspace solid phase micro-extraction and gas chromatography- mass spectrometry to analyse the volatile components, and then compare with varieties of chrysanthemums. That establish the specificity of volatile components in Shennong Xiangju perfectly; Then, bulid up the fingerprint of volatile oil in Shennong Xiangju, using principal component analysis to accomplish the comprehensive evaluation. And finally, based on the measured spectra with its antioxidant capacity studies, screened and identified the antioxidant active ingredient of Shennong Xiangju. 1, Using HS-SPME-GC-MS techniques to compare the volatile components of Shennong Xiangju with its original species and commercially popular varieties. Optimise solid phase microextraction head, powder dosage, solid phase micro extraction temperature and other single factor test to determine the optimum conditions for solid phase micro-extraction. Volatile compositions from five different chrysanthemum species are quite different, the commom ingredient are a total of only p-cymene, caryophyllene, farnesene. 2, Collecting 15 batches of Shennong Xiangju samples,the extraction of volatile oils is in accordance with the Chinese Pharmacopoeia 2010 edition of the first XD Appendix A method. Then caculated the oil yield of volatile oil. Using GC-MS for volatile analysis, and establish GC-MS fingerprints and evaluate intrinsic quality of different batches of volatile oil. 3, Through principal component analysis to reassemble the original data, making maps of the 23 indicators of the final by five main components to generalize the original information which including 88.966% indicators of the original amount of information. The first principal component(PC1) in Shennong Xiangju acts as the role of asthma indicators in volatile oil, the second principal component(PC2) as the analgesic effect of volatile oil in Shennong Xiangju, the third principal component(PC3) acts as the role of description to relieve cough. 4, Using the DPPH· method, optimize preferred antioxidant conditions in volatile oil of Shennong Xiangju. And on this basis, study of its antioxidant activity. The results showed that the ability to clear the DPPH· from strong to weak order is: BHT> Shennong Xiangju volatile oil> Vitamin E, and different batches of Shen Xiangju showed DPPH· clearance different. 5, On the basis of GC-MS fingerprint from Shennong Xiangju and its related pharmacodynamic studies, looking for "effective component group" contact effect with the antioxidant activity, access to 15 characteristic peak area from the GC-MS fingerprints of Shennong Xiangju oil and quantitative data of oxidation inhibition. Using gray relational analysis technique to measure the relative correlation, determine the chemical composition of fingerprinting represented on the efficacy of the size of the contribution. And then, according to the contribution of oxidation inhibition by each characteristic peak in GC-MS fingerprint of Shennong Xiangju oil, to speculate the extent of its antioxidant effect.The subject of the volatile components from oil in Shen Xiangju were systematic studied and combined with anti-oxidative activity. Thus to screen out activity peaks and anti-oxidative activity of the structure by knowing the spectrum-effect relationship. It is expected to be the new all-natural, safe antioxidant drugs and combining abundant resources in Hubei with local characteristics. Providing a scientific basis for the further development of Shennong Xiangju which have an important academic value and practical significance.
