Objective: This experiment takes invigorating spleen and stomach on behalf of Liu Junzi Tang as the research object,applied chemistry cancer-causing agents 4NQO to induce mouse model of esophageal cancer,observing Liu Junzi Tang to the model’s intervention for the quality of survival, lymphocyte subgroup cytokines, and comparing with on behalf of Qige San with regulating qi-flowing for eliminating phlegm effect for the treatment of esophageal cancer, to preliminary discussion on the mechanism of action of this method in the treatment of esophageal cancer, and provide theoretical basis for clinical application.Methods: 4NQO with 1, 2 propylene glycol mixture concentration of 2% of the mother liquor, then use clean water mixture concentration of 100 mu g/ml, through the induction of esophageal cancer in C57 BL / 6 mice model of drinking water, at the same time 6 gentleman in building group and Qi Ge mice simultaneously and independently drinking 6 gentleman soup and rev diaphragmatic payroll decoction for 16 weeks.Experiments are grouped into: A. normal control group: normal feed, free drinking pure water;B. model group: conventional feed, drink 100 mu g/ml 4NQO free drinking water in the dark the bottle;C1. 6 gentleman groups: 4NQO building started drinking 6 gentleman soup at the same time;C2. Liu Junzi Tang prevention group form the rev diaphragmatic scattered group is a bit thin, can activities.2.Weight changes: normal group 1-17 weeks weight increasing, after maintain at about 25 g. Model group 1-17 weeks of falling weight, maintain after about 17 g. Liu Junzi Tang Group 1-17 weeks constantly slow weight gain to about 23 g, and maintain to 23 weeks, then slow down. Qige San group of 1 to 17 weeks slow weight gain to about 25 g, decrease slightly later, weight at about 24 g.3. Diet :Water intake of normal group continued fluctuating around 4.3ml per day. Water intake of Model group、Liujunzi decoction group and qige powder group were droped Compared with the normal group after 16 days. Compared with the Liujunzi decoction group and qige powder group,water intake of model group were drop after 100 days. At the same time,compared with before,water intake of Liujunzi decoction group and qige powder group were steady had an upward trend. Food intake of normal group continued fluctuating around 3.2g per day. Compared with normal group,Food intake of model group,Liujunzi decoction group and qige powder group were lesser but had no significant difference.Compared with Liujunzi decoction group and qige powder group,Food intake of model group similar, Food intake had no significantly reduced.4. tissue in normal mice external form one eye can see smooth uniform, no bumps swelling;4NQO esophageal tissue in model mice esophagus overall inhomogeneous external form, has a protuberant swelling; Liu Junzi Tang and Qige San group of mice and normal mice esophagus surface are similar, not protuberant enlargement.Pathology showed normal esophageal mucosa epithelial basal cell layer、stratum spinosm、stratum corneum、submucosa and muscular layer in normal group; esophageal mucosa epithelial basal cell layer represented abnormal hyperplasia in model group and Liujunzi decoction group, but “cancer pearl” and “pathological division image” were only found in model group; basal cell layer occurred slight abnormal hyperplasia, stratum corneum liquefaction was almost disappeared in Qige powder group.5. Viscera weight and viscera index: model group kidney, spleen, heart, liver weight significantly lower than normal group, Liu Junzi and rev diaphragmatic powder can reduce 4NQO on the influence of the kidney, spleen, heart, liver weight, especially on the function of liver and spleen more obvious(P < 0.05), the effects on liver Qige scattered group is better than 6 gentleman decoction group(P < 0.05).Weight model group was obviously lower than other groups, Liu Junzi and Qige powder can significantly reduce the weight loss caused 4NQO(P < 0.05), and diaphragmatic function significantly better than Liu Junzi group(P < 0.05).Kidney, heart, lung index of model group was obviously greater than the normal group, Liu Junzi and Qige group(P < 0.05), Liu Junzi kidney, heart, lung index is greater than the normal group and Qige group(P < 0.05);Liver, spleen index in normal group and model group, Liu Junzi, Qige group has no difference(P > 0.05).6. Plasma cytokines: model group IL-12p70 、IL-6 and IFN-γ is significantly higher than normal control group(P < 0.05), TNF and MCP- 1 model group has a rising trend, but IL- 10 has a lower trend; Liu Junzi and Qige the 6 kinds of cytokines were scattered group was obviously higher than that of normal group(P < 0.05), Liu Junzi is better than Qige group(P < 0.05).7. Peripheral blood lymphocyte classification: CD3 + lymphocytes compared with control group, model group increased significantly(P < 0.05), CD19 + lymphocytes decreasing trend, increase trend, a proportion of CD3 + / CD19 + and CD3 + CD8 + cells increased, CD3 +, CD4 + / CD3 + CD8 + ratio decreases. Each index of Liu Junzi with diaphragmatic scattered and there is no difference between the normal group.Conclusion:1. 4NQO-induced esophageal mice model was successful, abnormal hyperplasia of basal cell layer was the main pathological change, “cancer pearl” and “pathological division image” were observed in model group. Compared with Liujunzi decoction, Qige powder showed better effect in prevention and cure of esophageal cancer.2. Liu Junzi Tang and Qige San can improve the quality of life of 4NQO induced C57 BL / 6 mouse model of esophageal cancer, effectively improve diet, weight loss caused by esophageal cancer, and can prevent and treat the pathological damage of esophageal tissue swelled swelling in mice and prevent 4NQO induced the formation of esophageal cancer.3. Liu Junzi Tang and Qige San can improve kidney, spleen, heart, liver, lungs and other organs weight and the influence of the corresponding viscera index of 4NQO induced C57 BL / 6 mouse model of esophageal cancer.4.Liu Junzi Tang and Qige San can increase the mice peripheral blood IL-12p70 、IL-6 、IFN-γ、TNF、MCP-1 and IL-10 cytokine levels.5. Liu Junzi Tang and Qige San can correct hum oral and cellular immunity obstacles in mice caused by 4NQO.
