Experimental Study Of Qingre Liangxue Qufeng Tongluo On Adjuvant Arthritis In Rats

ObjectiveRheumatoid arthritis is an autoimmune disease. Qingre Liangxue Qufeng Tongluo is a common method used in the treatment of rheumatoid arthritis,the research from literature and experimental study of Qingre Liangxue Qufeng Tongluo method on the mechanism of rheumatoid arthritis.In this experi ment, the adjuvant arthritis(AA) as the RA animal modol. Observation on T lymphocyte subsets in rats, AA related cytokines TNF-α、IL-1β、CD4+CD25+Tregs,To explore mechanisms of Qingre Liangxue Qufeng Tongluo method on this animal model.To provide the experimental basis for the clinical treatment of RA.Methods1.Theoretical research: Document analysis and induction of traditional Chinese medicine on the etiology,pathogenesis diagnosis and treatment of rheumatoid arthritis,explore the theoretical basis of experimental treatment for rheumatoid arthritis.2.Experimental study:①Making model and giving medicine:SD rats were divided into blank group, model group, comparative group(Wang Bi Granule) and experimental group( empirical formula).According to the document’s method,except the blank group,The other groups in the rat’s right plantar subcutaneous inject with the complete Freund’s adjuvant0.15 ml.The blank group with the same method, injecting with saline 0.15 ml.After 7 days,the rats of comparative group were treated with 1.88g/(kg·d) Wang Bi Granule and experimental group were treated with 29g/(kg·d) empirical formula.The blank group and model were treated with physiological saline 3 ml/d.②Observing the AA rats’ ordinary circumstances, measuring the weight of rats, paw thickness, assessment of arthritis index。③Observing on T lymphocyte subsets in rats, AA related cytokines TNF-α、IL-1β、CD4+CD25+Tregs.The data were analyzed by SPSS17.0 statistical software.Results1.Theoretical research:Deficiency in origin and excess in superficiality,cold,hot,mixed,mixed stasis-heat blockage for the pathogenesis of RA main characteristics.With the development of disease in remission stage, “ill will and blood stasis”, “long illness into complex” disease gradually emerged of the actual situation in sight,cold,hot,mixed,repeatedly in the pathogens under produce phlegm,blood stasis and the pathological products such as heat,viscera,hurting the qi of yin and yang,shown as attack lingering hard to recover.Therefore,based on this use of qingre liangxue qufeng tongluo therapy for rheumatoid arthritis.2.Experimental study:①The ordinary situation of the rats:Except the blank group,the rats’ foot appeared swelling after 6h modeling.Reached a peak after 24 h,lasting 3 days,rediscovered swelling after 7-8 days, influencing knee joint,skin appears ulcer,licking foot,claudication,drag phenomenon.Low reaction to outside stimuli.Diet and activity reduced, diarrhea,hair become dry.The secondary lesion occurs in 12 days after the inflammation,the symptoms tend to get worse progressively.Compared with the blank group, the model group rats appeared the above performance.The experimental group and Wang bi granules group general overall better than the model group.②The performance of changes on weight, each rat paw swelling and arthritis index:No significant difference in the mean weight in the same treatment stage of rats.The rats after modeling to the end of the treatment process of weight is an overall upward trend.Each rat paw swelling in the same phase of treatment compared with the blank group, there was statistical significance(P<0. 05).After 2 weeks of treatment, the experimental group signifycantly relieve swelling, and significant different compared with model group.Compared with the blank group the rats showed different degrees of joint inflammation after 7 days,the difference has statistical significance(P<0.05).③The performance of rat’s peripheral blood T lymphocyte subsets and the changes of CD4+/CD8+ :Compared with the blank group, model group had significant difference in CD4+ level, CD8+ level, and the ratio of CD4+/CD8+.The experimental group can decrease the level of CD4+, compared with the model group, the difference was significant(P<0.01).The experimental group increased the level of CD8+,compared with the model group, the difference was significant(P<0.05).The experimental group can decrease the ratio of CD4+/CD8+, compared with the model group, the difference was signi-ficant(P<0.01).④Changes of peripheral blood serum of rats in the cell factor TNF-α、IL-1β:The model group TNF-α, IL-1β cell levels were elevated,compared with the blank group, the difference was significant(P<0.01).The experimental group can decrease cell factor TNF-α,IL-1β level,compared with the model group,the difference was significant(P<0.01).⑤The rats’ peripheral blood CD4+CD25+Tregs cell changes:CD4+ CD25+Tregs in model group was decreased, compared with the blank group, there was significant difference(P<0.01).Traditional Chinese medicine can increase the level of CD4+CD25+Tregs cells, compared with model group, the difference was significant(P<0.01).Conclusions1.Qingre Liangxue Qufeng Tongluo method can improve AA rats’ general condition,inhibit AA rat paw swelling and arthritis index score,and control the development of inflammation,alleviate the pathological damage induced arthritis in rats.2.Qingre Liangxue Qufeng Tongluo method can decrease the level of CD4+ lymphocytes,increase the level of CD8+ lymphocyte,decrease the ratio of CD4+/CD8+,which regulate T lymphocyte subsets by two-way.It may be the main role is effective in the treatment of rheumatoid arthritis mechanism.3.Qingre Liangxue Qufeng Tongluo method can reduce the cytokines in rats,inhibit the secretion of TNF-α、IL-β.It may another important mechanism which is effective in the treatment of rheumatoid arthritis.4.Qingre Liangxue Qufeng Tongluo method can increase the level of CD4+CD25+Tregs cells in AA rats,and achieves the prevention of rheumatoid arthritis objective, which may be the effective treatment of rheumatoid arthritis and an important mechanism.
