Insomnia, has so many other names in china, like “eyes cann’t closed”、“cann’t go to sleep” 、“cann’t lie on the bed” etc. western medicine often referred it as insomnia, which means having trouble to fall asleep,or to maintain sleep(including easy to wake up, go to bed early and hard to fall asleep when bothered), that leads to the decline in the quality and quantity,the reducing can not satisfied the body’s physiological needs, significantly affect daily social functioning and quality of life. This paper absorbed the mentor’s former researc h, then continue to explore insomnia model’s establishment and observe chinese medcine’s function and the mechanism.Theoretical research: "Yellow Emperor" thinks that there exist a complex relationship between “Shen” and sleep, “Shen” dominates all activities of the living body; the change of yin and yang, the circulation of nutrient qi and defensive qi, the rise and fall of “qiaomai”, thy all closely related to the body’s sleep. nutrient qi and defensive qi are the foundation of soul activities, whose circulation and the rule of “yin yang” are the key point of sleep activities,The circadian of nutrient qi and defensive qi and” yinyang”,the regulation of their pace also ensures physiological self-regulation of sleep. Insomnia has so many etiologies, and pathological mechanism is very complexing, other deficiency or excess, deficiency of heart and liver yin, deficiency of heart and gallbladder qi, disharmony between heart and kidney, liver stagnation and qi stagnation that leads to disturbing “xinshen”, stagnated gallbladder qi with disturbing phlegm transforming into fire likely to disturb heart, hyperactivity of heart fire, et al. they all can lead to insomnia, and many of them have symptoms of irritable.The treatment should also identify the etiology and pathogenesis, and treatment based on syndrome differentiation.Experimental study: Methods: First through the model selection method to select the appropriate sleep deprivation model in mice, then doing the research of three grass’ s Hypnogenetic Pharmacodynamics Study, then use the touch overlay for-cage method as a method of sleep deprivation in mice based on the results, choose three grass as intervention medication. Adaptive feeding those mice, then doing the water maze test to select those normal and heathy ones, about 60 mice that were divided into five groups, each group have 12, Groups were: control group, model group, comparison group, three grass sedative high dose group, three grass sedative side low dose group. During the adaptive feeding process, maintain they were in a single cage, light: dark = 12h: 12 h, control indoor temperature maintained(22 + 2) ℃. Weighing, recording, lavaging every day, mice were observed daily for recording weight, movement behavior, mental health, fur gloss, the amount of drinking water and pooh et al. Modeling.using the Morris water maze test to detect the changing of learning and memory ability of mice.the using of autonomous activity box experiment to detect mice’s movements and change of activities.Using histochemical method to test each set of experiments’ s free radicals(SOD, MDA) and NO, NOS and the level of Ch AT、TCh E in brain tissue of miceResults: In the autonomous activity test, compared with the blank group, mice in combined group, gentle handling group’s event number and the total distance were significantly decreased(P < 0.05);cage changing group have the tendence to decrease, but the difference was not significant. In the water maze test capability, compared with the blank group, the other 3 group’s mice have a longer latency time and swim longer(P<0.01,P<0.05), The test of ability to detect in space exploration, The results are consistent with the previous experimental results, compared with the blank group, the other 3 groups have a less time to cross the platform, spent less time in section one and spent more time to find the platform(P<0.01,P<0.05), In promoting sleep experiment, the results show that when giving enough medicine of three grass, creature can sleep well. After sleep deprivation in mice, general indicators to be worse than the blank group of mice, the amount of weight gain are lower than the blank group of mice, but the western medicine group and three grass sedative side high dose group can improve this situation. In the water maze test, three grass sedative side in the intervention group and the target number of mice through the platform quadrant time increased significantly, 1st arrival time was significantly shortened, the difference was significant different(P<0.01, P<0.05). After three grass sedative side interventions mice’s SOD activity increased(P<0.05), MDA content decreased(P<0.05), NO and NOS content decreased(P<0.05), Tch E activity and Ch AT activity was significantly higher(P<0.05)Conclusion: 1. The combination of gentle handling and cage changing method is a safe, reliable, feasible and economical sleep deprivation method.2. The medicine of three grass has a significant effect on Sedation and hypnosis, especially the higher doze.3. The medicine of three grass can improve the quality of mice’s sleep situation, and hence the ability of learning and studying.
