| [Objective]This study uses the multi-factor synthesis under the action of the rat model with chronic atrophic gastritis to definite operation objectively the curative effect of particles on the chronic atrophic gastritis rats, and discuss its mechanism of action, and provides the basis of clinical and experimental research, which will be the foundation of developing Zhi Zhu granula.[Methods]1. Establishing and improving the rat model with chronic atrophic gastritis, comparative study on the biological characteristics and the pathology.2. Through the definite particles of high, medium and low dose effects on chronic atrophic gastritis rats model, compare the differences in weight of rats and repairing gastric mucosal, then searching for optimal dosing concentration.3. Detecting the protein expression of TFF1, COX-2 and VEGF by Western Blot and immunohistochemical technique.4. Detecting the mRNA expression of TFF1, COX-2 and VEGF by Realtime-PCR technique.[Results]1. The weight of the CAG model rats obviously lower than normal group, difference is statistically significant (P<0.01); The growth rate of the CAG model rats obviously lower than normal weight group, difference is statistically significant (P<0.01); The stomach tissue pathological of CAG model rats compared with normal group have obvious inflammatory cells infiltration, erosion and glands atrophy.2. All the rats weight growth rate of treatment groups were higher than that of the CAG model group, difference is statistically significant (P<0.05). The weight growth rate of Zhi Zhu granula groups were higher than that of positive medicine treatment group, difference is statistically significant (P<0.05).3. TFF1 protein expression of the CAG model rats is lower than the normal group, the dose groups were obviously higher than that of positive medicine group, difference is statistically significant (P<0.01); The cox-2, VEGF protein expression of CAG model rats was higher than normal group, cox-2, VEGF protein expression of the Zhi Zhu granula groups is lower than the model group, and the high-dose group compare with the positive medicine group has no statistical difference (P>0.05).4. TFF1 mRNA expression of the CAG model rats is lower than the normal group,the high-dose and middle-dose groups were obviously higher than that of positive medicine group,difference is statistically significant (P<0.01); The COX-2, VEGF mRNA expression of CAG model rats was higher than normal group, COX-2, VEGF protein expression of the Zhi Zhu granula groups is lower than the model group, and the high-dose group is lower than the positive medicine group, the difference has no statistical difference (P>0.05).[Conclusion]1. Through the examination of rat gastric histopathological, it can confirm that deoxycholic acid sodium, ammonia and hunger disorder successfully conducted in rats with chronic atrophic gastritis model.2. The Zhi Zhu granula particle therapy can increase the growth of the body weight of rats, and can improve stomach tissue pathological morphology.3. The Zhi Zhu granula particles can increase the expression of TFF1 which is the gastric mucosal protection factor of CAG model, and can cut the expression of cox-2 and VEGFwhich are the gastric mucosal damage factort. |
