Objective:To through the experiental research of Dihuang Tusi Tiaojing Keli, observe the change of it to the sex hormone of premature ovarian failure(POF) rats and pathological ovarian tissue morphology, the effect of the protein expression of the stem cell factor (SCF) and insulin-like growth factor-I(IGF-I),the expression of gonadotropin releasing hormone receptor messenger RNA nucleotides (GnRHR-mRNA) to investigate the mechanism of the prevention and treatment of Dihuang Tusi Tiaojing(DHTSTJ)KeLi on rat model with POF,which provide ideas For clinical applications of the POF with Traditional Chinese medicine caused by the immune inhibitors.Methods:The 60 female adult SD rats with regular estrus were randomly divided into five groups:blank group, model group and western medicine, Chinese medicine group and prevention group, gave them inject 60 mg/kg cyclophosphamide one time to make premature ovarian failure (POF)rats models. After 8 weeks, observe the rats oestrous cycle with the vaginal exfoliated cells smear method and the ovarian tissue morphology changes of the rats through the HE staining,test the level of the rats’estradiol and follicle stimulating hormone with chemiluminescence method, look into the protein expression of SCF and IGF-I with the immunohistoche mistry method and test the expression of GnRHR-mRNA of the rats’ovarian through Semi-quantitative RT-PCR.Result:(1)compared with the model group, the other groups reduce the level of FSH and increase the level of E2 obviously (p<0.05).compared with the estrogen control group, The prevention group is more obvious (p<0.05).(2) there was obviously atrophy and even fibrosis of the model group’ovarian, the follicles at all levels were decreased significantly and corpus luteum degraded, while atresia follicles increased under the light microscope.Compared with model group, the organizational structure of ovarian was clear, the follicles at different levels and the mature follicles, the number of corpus luteums increased in the other groups.Compared with western medicine group, the prevention were significantly improve the ovarian tissue structure,and increased the number of follicles.(3)Compared with the model group, the protein expression of IGF-I in the other groups increased (p<0.05) and the expression of SCF decreased (p<0.05). While compared with the estrogen control group, the prevention group is more obvious (p<0.05)(4)Compared with the model group, the expression of GnRHR-mRNA in the other groups increased(p<0.05). While compared with the estrogen control group, the prevention group is more obvious (p<0.05)Conclusion:(1) Dihuang Tusi Tiaojing Keli can reduce the level of FSH and increase the level of E2 in the premature ovarian failure rats,increase the number of mature follicle to improve the ovarian function. (2) Dihuang Tusi Tiaojing Keli may regulate the expression of IGF-I in granular cells and the GnRHR-mRNA to treat the premature ovarian failure.(3) Dihuang Tusi Tiaojing Keli may regulatethe expression of SCF in granular cells to protect the ovatian and reduce the injury bycyciopnospnamide.
