Objective: Through the experimental research of Dihuang Tusi Tiaojing Keli,observe the change of it to the secretion of estrogen of premature ovarianfailure(POF) rats and pathological ovarian tissue morphology, the effect of theprotein expression of gonadotropin releasing hormone receptor (GnRHR) andthe expression stem cell factor (SCF)mRNA to investigate the protective effectof Dihuang Tusi Tiaojing Keli to the premature ovarian failure rats induced bycyclophosphamide.Methods: Inject60mg/kg cyclophosphamide one time to make prematureovarian failure (POF)rats models and separate them into5groups: the blankcontrol group,the model group,the estrogen control group, the prevention groupof Dihuang Tusi Tiaojing Keli (the prevention group)and the traditionalChinese medicine group of Rehmannia Tusi Tiaojing Keli(the traditionalChinese medicine). After8weeks, test the level of the rats’estradiol, folliclestimulating hormone and luteinizing hormone with chemiluminescence method,observe the pathological changes of the ovary of the rats under the lightmicroscope,look into the expression of GnRHR with the immunohistochemistry method and test the expression of SCF-mRNA of the rats’ ovarianthrough Semi-quantitative RT-PCR.Result:Compared with the blank control group, the peripheral blood of POFrats in the model group have much higher level of FSH and LH(P<0.05) andmuch lower level of E2(P<0.05); compared with the model group, theprevention group, the traditional Chinese medicine and the estrogen controlgroup can reduce the level of FSH and LH of the peripheral blood of POF ratsobviously and increase the level of E2. The prevention group is much clearer(P<0.05).Dihuang Tusi Tiaojing Keli can obviously expand the number of growingfollicles and corpus luteum in ovarian tissue, reduce locking cells significantlywith more mature follicle and less ovarian interstitial lymphocytic infiltration. Dihuang Tusi Tiaojing Keli can increase the expression of granular cellGnRHR. Compared with the model group, the other groups increased (P<0.05) while compared with the estrogen control group, the prevention groupincreased(P<0.05). Dihuang Tusi Tiaojing Keli can decrease the expression ofgranular cell SCF-mRNA.Compared with the model group, the other groupsdecreased(P<0.05)while compared with the estrogen control group and thetraditional Chinese medicine, the prevention group decrease(P<0.05).Conclusion:Dihuang Tusi Tiaojing Keli can reduce the level of FSH and LH ofthe premature ovarian failure rats, increase the level of E2and the expression ofgranular cell GnRHR, decrease the level expression of granular cellSCF-mRNA, protect the ovatian, and at the same time, reduce the ovarianinjury of the use of cyclophosphamide.
