Objective:In vivo and vitro research, IFN-y anti liver fibrosis and lung injury fibrosis has been confirmed.The study aimed to observe the effect of IFN-γ on both the mRNA and protein levels of NRK-49F cells and investigate the potential mechanism of renal interstitial fibrosis.Methods:NRK-49F cells were cultured in vitro, and treated with 1000U/ml,2000U/ml, 3000U/ml,4000U/ml,5000U/ml concentration of IFN-y, repectively. Control group was treated with 0.9% physiological saline.The proliferation of NRK-49F cells was assessed by MTT colorimetric method. Both the MCP-1 mRNA and protein levels were tested by RT-PCR and Western-blot, respectively.Results:1. NRK-49F cells proliferation:Used single factor analysis of variance,the mean OD levels of different concentration of IFN-y group were significantly higher than that of the control group (p<0.05). IFN-y promoted the proliferation of NRK-49F cells in a time-dependent manner (p<0.05), but not in a concentration-dependent manner (p>0.05).1. The expression of MCP-1 mRNA:Compared with the control group, the MCP-1 mRNA levels of different concentration of IFN-y group were significantly increased (p<0.05). There is no significant difference in the expression of MCP-1 mRNA levels (P>0.05).2. The expression of MCP-1 protein:Compared with the control group, the MCP-1 protein levels of different concentration of IFN-y group were significantly increased (p<0.05). There is no significant difference in the expression of MCP-1 protein levels (P>0.05). Conclusion:IFN-y promoted the proliferation of NRK-49F cells in a time-dependent manner. At the genetic level, IFN-gamma could upregulate the expression of MCP-1 mRNA levels in NRK-49F cells. IFN-y could promote secretion of MCP-1 protein in NRK-49F cells. IFN-y promotes renal interstitial fibrosis via increasing MCP-1 levels.
