Curculigo orchioides is the category of perennial herbs which has high me dicinal value.With the number of natural population of Curculigo plant has decl ined and the growth of harsh environment, leding the quality of medicinal plan t decling.Curculigoside is the standard of evaluation in Chinese medicine qualit y. Therefore, it improve the rate of utilization in effective ingredients of traditiona 1 Chinese medicine,and it is helpful for the evaluation of Chinese medicine extr action plant medicinal materials quality and the relatively natural products provid e practical significance by the extraction process of Curculigoside reasonabliy an d purificational experimental design method. The main results are as follows:The optimum conditions for the study of reflux extraction by response surface methodology from Curculigoside were:the concentration of ethanol of 76.09%, the extraction time of 118.65, the olid-fluid ratio of 13.73. Consid ering the actual condition, it can be adjusted to the concentration of ethanol of 76%, the extraction time of 120, the olid-fluid ratio of 1:14.Based on experiments of traditional reflux method, ultrasonic extraction combi nedwith modern technology to optimize the extraction process of Curculigoside.Thr ough the single factor experiment and the response surface experimental design an d Box-Benhnken analysis to determine the Curculigoside ultrasound assisted reflu x optimum technological parameters of extracting experiment were ultrasonic ou tput power of 326.04W, the concentration of ethanol of 80.8%, the olid- fl uid ratio of 9.33. Considering the actual condition, it can be adjusted to the ul trasonic output power of 320W, the concentration of ethanol of 81%, the ol id-fluid ratio of 9.5.Using the D101 macroporous resin for purification of Curculigoside, investigat ion of D101 macroporous adsorption resin of Curculigoside in static and dynamic adsorption and desorptioncapacity.Optimization purification condition were as fol lows:the time of absorption was 8h,pH of sample liquid was 6,pH of ethanol was 8,temperature of absorption and desorption can be room temperature, conce ntration of Curculigoside in sample liquid was 0.16 mg/ml, flow rate was 2 B v/h, eluent concentration was 30%ethanol and the dosage was 8 Bv.
