Purpose:To investigate, the effects and mechanism of cyanidin-3-glucoside (C3G) on human cancer of bile duct QBC939 cell line.Methods:After human cancer of bile duct QBC939 cell line was treated with C3G,the HIF-1a protein was determined by immunofluorescene technology,and cellular proliferation was detected by MTT assay.The apoptosis ratio was detected by flow cytometry.The expression of HIF-1a protein was determined by Western blotting assay and immunofluorescene staining methods.The expression of mRNA both of HIF-1a and its downstream target gene of EPO,VEGF was determined by RT-PCR.Results:Compared with the control group,the activity of HIF-1a protein of QBC939 cell was significantly inhibited by C3G,and presented concentration dependent manner.The proliferation of QBC939 cells was significantly inhibited by C3G,but there was no obviously cytotoxicity,and C3G was markedly induced the apoptosis.The expression of HIF-1a protein and downstream target genes mRNA of EPO and VEGF were significantly down-regulated after treatment of C3G in QBC939 cells,and presented concentration dependent manner.Conclusion:C3G can significantly inhibits the proliferation of QBC939 cells, the mechanism of apoptosis may be related to inhibition the activity of HIF-1a factor in QBC939 cells.
