[Object]Rhizoma paridis, which chemical constituents are complex. Polyphyllin Ⅰ (PP Ⅰ) is one pharmacological active substance extract from it. Clinical and experimental study has obtained good results especially in tumors. The aim of this study is to explore the effects of the polyphyllin I on the invitro anti-proliferation of human melanoma cell lines of A2058 and G361.[Method]l.By addition of different concentrations of polyphyllin I dealing with human melanoma cell lines of A2058 and G361 at different times. MTS assay was used to exam the status of cell proliferation.2. Add to polyphylla I in cultured melanoma cells and observe the differences in cell morphology at the optical microscope.3.By culturing of melanoma cells with polyphyllin I and detected apoptosis by flow cytometry(FCM).[Result]1.It was observed that polyphyllin I could inhibit the proliferation of the human melanoma cell lines of A2058 and G361 within the dosage from 0.625μg/ml to 10μg/ml and the effects were in a significant dose-dependent manner, It could induce apoptosis of the melanoma cell lines.2. Difference in cell morphology were observed under optical microscope.3. By FCM, apparent apoptosis could be observed at the concentration of 2.5μg/ml at 12h,24h and 48h:the longer the time, the higher the rate of apoptosis. And use different concentrations of 1.25μg/ml,2.5μg/ml,5μg/ml culturing cells for 24h, shows dose-dependent apoptosis.[Conclusion]Polyphyllin I can inhibit the proliferation of human melanoma cell lines and induce apoptosis.
