Objective: Melanoma is one of the most common cancers in the dermatology department.Its morbidity ranks third among skin cancers,while mortality tops the list.The treatments of melanoma include surgical treatment,chemotherapy,radiotherapy,immunotherapy,personalized biotherapy and individual target therapy.Though these therapies have certain positive effects on patients,there still exist many problems including poor efficacy and great side effects.Therefore,it is important to develop some new and effective drugs to prevent and treat the melanoma.At present,some natural plant extracts show unique advantages in anti-tumour field.Fucoidan can be extracted from brown algae or invertebrate with various biological activities.A large number of researches found that fucoidan has certain therapeutical cancers of lung,colon,breast,stomach and liver.Meanwhile,fucoidan not only can promote the apoptosis of tumor cells through various mechanisms,but cooperate with some medicines to increase the sensitivity and efficacy of chemotherapy drugs,and reduce their toxicity.Based on the cell experiments in vitro,some foreign scholars found that fucoidan can inhibit B16 proliferation and promote apoptosis.At present,there are no relevant researches about the effects of fucoidan on melanoma cells in China.This research studied the effects of fucoidan on the proliferation and apoptosis of melanoma A375 cells as well as the Caspase-3 protein expression via in vitro cell experiment, aiming to provide theoretical and experimental foundation for the clinical application of fucoidan on preventing and treating melanoma. Methods: A375 melanoma cell cells were cultured in vitro. Then CCK8 method was applied to detect the cell proliferation, calculate the inhibition of cell proliferation, and draw the dose-effect curve to obtain half of proliferation inhibition concentration(IC50) after intervening in the cells via fucoidan with different concentrations for 24 hours. Furthermore, Annexin V-FITC/PI double-staining method and flow cytometry(FCM) were used to observe the effect on the cell apoptosis; the western blot method was adopted to detect the changes of Caspase-3 apoptotic protein expression Then, a preliminary discussion could be held about effects of fucoidan on cell proliferation and apoptosis as well as its relevant mechanism. Results: Compared with the control group, CCK8 showed that the inhibition of A37 cell proliferation was increased with the rising drug concentration after 24 hours of fucoidan. Half of proliferation inhibition concentration was calculated as(IC50)0.54mg/ml; the flow cytometry detection results demonstrated that the cell apoptosis rate increased obviously after the fucoidan; the Western blot results indicated that the Caspase-3 expression was increased with the rising fucoidan concentration. Conclusion: Fucoidan can inhibit the proliferation and accelerate the apoptosis of A375 cells.
