Study On The Effect Of Huagan Tongluo Fang Towards A Murine Model Of Primary Biliary Cirrhosis

Objective:To explore the prevention and treatment effect by Huagan Tongluo formula on a murine model of primary biliary cirrhosis (PBC).Methods:Murine models of PBC developed by injection of polyinosinic polycytidylic acids (poly I:C) in C57BL/6 mice were treated with Huagan Tongluo formula granule concentrated solution (high dosage with 14.72g/ml and middle dosage with 3.68g/ml), vehicle by gavage respectively. After 20 weeks’treatment, the mice were killed, sampled, detected while the differences among all groups were analyzed and compared. Take serum to detect the liver biochemical indexes. The hepatic pathology morphologic changes were observed by HE staining with partial liver. To build animal models in the same way, then randomly divided into 7 groups:blank control group, model group, ursodeoxycholic acid group(UDCA 91.67mg/kg/d), Huagan Tongluo formula granule concentrated solution group(middle dosage with 7.36g/ml and low dosage with 3.68g/ml), combine traditional chinese and western medicine treatment group(combine UDCA and Huagan Tongluo formula). All the groups were continuous dosing 8 weeks respectively. After the mice were killed, sampled, detected while the differences among all groups were analyzed and compared. Take serum to detect the liver biochemical indexes. The hepatic pathology morphologic changes were observed by HE staining with partial liver. The expressions of a-Ⅰ collagen were detected with real-time quantitative PCR detecting system.Results:(1)AST and ALT indexes in groups given Huagan Tongluo formula and UDCA ameliorate more significantly than PBC model group, which had a statistically significant difference(P<0.05). Both Huagan Tongluo formula and UDCA can reduce the mice serum AMA/M2 high degree of expression. The changes of liver pathological tissues in groups with Huagan Tongluo formula improved more significantly than that of PBC model group. (2)Both Huagan Tongluo formula and UDCA can reduce a-I collagen content in liver tissue. (3)There was no statistical significance between traditional Chinese medicine treatment groups and UDCA group(P>0.05). (4)Combined drug group also showed good therapeutic effect, but the result less than traditional Chinese medicine and UDCA treatment groups.Conclusion:1) Huagan Tongluo formula can reduce the liver biochemical index and ameliorate the liver lesions in mice, which has a role in protecting the PBC mice liver and delaying of PBC’s development.2) Huagan Tongluo formula can be used as treatment of PBC.3)Combine traditional Chinese and western medicine treatment method does not show the advantage than ursodeoxycholic acid alone in the treatment of PBC.
