The Reserch Of Plasma Adiponectin SNP + 276 G/T 、 Leptin SNP- 2548 G/A Loci Gene Polymorphism In The Pilots

Objective:To discuss the plasma adiponectin 、Leptin levels in the pilots, general physical examination and comparison of any differences between the patients with coronary heart disease(CHD).And analyze the APN SNP + 276 G/T(rs1501299) loci、Leptin SNPS- 2548 G/A(rs7799039) loci gene polymorphisms have any obvious difference.Methods:Selection aged between 20 ~ 60 of 727 cases of the research object,Divided intothe flying group(473 cases) and general physical examination(200 cases),coronary heart disease group(54 cases) three groups, the flying group again according to the model group was divided into combat units(250 cases) and non-combat units(223 cases) in the two groups.Through the questionnaire survey and the basic measure for all participants’ general clinical information collection,such as age、height、weight. for the special group of the pilots,add to flight models,etc.All the subjects on an empty stomach after 12 h, The next day at 6:30 ~ 7:30 Venous blood was collected 4 ml,Using purple containing EDTA anticoagulant tube collect venous blood,drain out the 2 ml venous blood, 3500 r/min, the centrifugal 3 min, absorbs the serum used for determination of serum adiponectin and leptin. The remaining 2 ml blood diluted with 0.9% physiological saline blending,using pipetting device slow drop into the 10 ml centrifuge tube that it is equipped with 2 ml of lymphocyte separation medium, 3500 r/min,the centrifugal 3 min, the separation of the WBC layer, use pipettes absorb the WBC transferred to 1.5 ml EP tube,using 12000 r high-speed centrifuge for 1 min,abandon purify precipitation of all on clearliquid reserve with white blood cells, stored in-80℃ refrigerator,unified for extracting peripheral blood leukocyte DNA. Application of Elisa method to detect serum APN,Leptin concentration of three groups of people,and whole blood genomic DNA extraction,Taqman probe method is used to detect all the research object APN SNP + 276 G/T(rs1501299)、Leptin- 2548 G/A(rs7799039) loci gene polymorphsim. Analysis of the pilot group、general physical examination group、 coronary heart disease group and three groups of plasma adiponectin and Leptin levels of change,and detection of APN SNP +276 G/T、Leptin- 2548 G/A loci of different genotyping and allele differences between groups. And analysis APN SNP+276 G/T、Leptin-2548 G/A loci of different genotyping and its corresponding plasma levels have any difference between in groups,accordingto the flight group is divided into different models.Results:(1) Age、BMI in comparison between groups,there was no significant difference(P >0.05).(2)comparison APN plasma levels in three groups,coronary heart disease group(5.07±0.96ng/ml) is lower than the pilots group(4.61±1.34ng/ml), the pilots group is lower than general physical examination group(6.11±1.57ng/ml),there was a striking different(P<0.05);To compare the plasma levels of Leptin in three groups,coronary heart disease group(343.40±104.33pg/ml)is significantly higher than general physical examination group, and general physical examination group(325.59±58.08pg/ml) is higher than the pilot group(282.62±42.29pg/ml)(all P <0.05);(3) APN SNP +276 G/T loci、 Leptin-2548 G/A loci of each genotype frequency and allele frequency in the pilot group、general physical examination group and coronary heart disease group,there was no difference in the three groups(P>0.05).(4) combat units APN SNP+276G/T of GG、GT、TT in the three genotypes,APN plasma levels compared with non combat units,there was no significant difference(P >0.05). The AA、AG、GG three genotypes of combat units leptin-2548 G/A loci and its corresponding plasma Leptin level compared with the non-combat units,there was no difference(P > 0.05).Conclusion:(1) plasma adiponectin level of the pilot group is lower than generalphysical examination,indicates the pilot have more stress reaction state than general physical examination;⑵plasma leptin level of the pilot group is lower than general physical examination,it can’t prompt the pilot more inflammation reaction than general physical examination. ⑶ adiponectin gene SNP +276 G/T、leptin gene SNP-2548 G/A loci gene polymorphism has nothing to do with coronary heart disease. ⑷adiponectin gene SNP +276G/T loci 、 leptin gene SNP-2548G/A loci of genotypes and its corresponding changes of plasma adiponectin and Leptin levels have nothing to do with the flight model.
