Objective:for more than 30 years,in order to establish an ideal spinal cord ischemia reperfusion injury model,which with simple operations, less complications and high similarity, a large number of researchers tried to use more kinds of animals,different incisions and more kinds of blocking methods. But so far there is still no "gold" standard to create an ideal model with high clinical similarity, less complications or no complications. In addition, in some respects there is still a huge controversy. This study by comparing Zivin’s method to establish the rat and rabbit spinal cord ischemia-reperfusion injury model success rate, the common reasons for failure and improvement measures are discussed.Methods: using Zivin’s method(reperfusion after clipping the ventral aorta caudal to the left renal artery), we make 30 cases of adult New Zealand rabbits and SD rats spinal cord ischemia-reperfusion injury model respectively.All the New Zealand rabbits were divided into group A,B and C randomly.The ischemia time limit for the three groups is 30 minutes, 45 minutes and 60 minutes.They stand for mild, moderate and severe spinal cord ischemia-reperfusion injury. All the SD rats were divided into group a,b and c randomly.The ischemia time limit for the three groups is 30 minutes, 45 minutes and 60 minutes.They also stand for mild, moderate and severe spinal cord ischemia-reperfusion injury.We modified Tarlov’s criteria to evaluate the New Zealand rabbits’ and rats’ neuroethology changes after reperfusion.Compare the Tarlov score of group A and group a, group B and group b, group C and group c,using statistical mothed.Then compare the success rate of rabbit and rat model and analyze the reason, summarizes the methods to improve success rate of modeling.Result: In mild ischemia reperfusion injury models, The modified Tarlov score of mild ischemia reperfusion injury models of New Zealand rabbits and SD rats,after 6h, 24 h and 48 h are statistical different(P<0.05). In moderate ischemia reperfusion injury models, The modified Tarlov score of ischemia reperfusion injury models of New Zealand rabbits and SD rats,after 6h, 24 h and 48 h are statistical different(P<0.05).In severe ischemia reperfusion injury models, the modified Tarlov score of ischemia reperfusion injury models of New Zealand rabbits and SD rats after 6h, 24 h and 48 h are statistical different(P<0.05).The New Zealand rabbit groups successfully established spinal cord ischemia reperfusion injury model in 28 cases(93.3% success rate). The SD rat groups successfully established spinal cord ischemia reperfusion injury model in 3 cases(10% success rate).Conclusion: There is a big difference between the success rate when using New Zealand rabbits and SD rats to built spinal cord ischemia reperfusion injury models.The success rate when using SD rats is low Anatomical differences may be the main reason of the different success rate of Zivin’s method to establish the New Zealand rabbit and SD rat spinal cord ischemia reperfusion injury models.
