Objective:To discuss the depressurization of tetramethylpyrazine in spontaneously hypertensive rat, and detect oxidation reaction, inflammatory, NO and iNOS to find out the mechanism of tetramethylpyrazine in spontaneously hypertension.Methods:Systolic blood pressure in3-weeks,16-weeks SHR rats and WKY rats were detected and serum cholesterol and oxidation reaction were also detected to find out the success of spontaneously hypertensive rat model.10WKY rats and50SHP rats were divided into normal, positive control and three experimental groups with different dosage, and captopril group. Normal group and positive control group were suffered with saline, three experimental groups were suffered with tetramaethylpyrzine of15mg/kg,30mg/kg, and60mg/kg. Captopril group were suffered with captopril of lmg/kg/d for4weeks. Before administration, all rats were detected systolic blood pressure, and systolic blood pressures were also detected at7th,14th,21st, and28th day. Last administration, serum of all groups were collected, and heart, liver tissue were collected, stored in-80℃.Results:1. There were no significantly difference of SBP, LDL-C and HDL-C levels in3-weeks and16-weeks WKY.2. GSH, CAT and SOD activities in16-weeks SHR rats were significantly lower than these in16-weeks WKY rats, while MDA significantly decreased.3. NO and iNOS levels in SHR rats were similar with these in WKY rats.4. Systolic blood pressures in SHR+CXQ15, SHR+CXQ30, SHR+CXQ60and SHR+CAP groups were significantly decreased than SHP group with administration of tetramethylpyrazine.5. GSH, CAT and SOD activities in SHR+CXQ15, SHR+CXQ30, SHR+CXQ60and SHR+CAP groups were significantly increased than SHP group with administration of tetramethylpyrazine, while MDA decreased.Conclusion Tetramethylpyrazine showed significantly depressurization effect on systlic blood pressure of SHP rats, which might via anti-oxidation, decreasing inflammatory factors and increasing NO or iNOS levels.
