Breast cancer is one of the most common malignant tumors in women and serious harmto human health. In recent years, with the improvement of living standards, breast cancer becamethe main reason of death in women. Patients with new onset are nearly one million. The trend ofyounger onset age is more and more serious around the world each year. So cancer earlydetection and diagnosis is important for tumor cure rate. Circulating tumor cells existed inpatients with peripheral blood is a major cause leading to tumor metastasis and recurrence.Detection of circulating tumor cells has important significance for discovering and preventingthe occurrence of tumor. The circulating tumor cells can be stranded somewhere in the body. It iseasy to form lesions under the condition of the environment for the growth. The traditionaldetection tumor treatment has significant side effects. Thus a fast, efficient and small damagedetection treatment is particularly necessary.Circulating tumor cells can be effective to capture by indwelling needle. The capture ofcirculating tumor cells doesn’t need to take extra blood which is the advantage of this approach.The method can be completed in the process of transfusion, has less damage and high sensitivity,can be accepted easily. The detected nano probe of two kinds of estrogen receptor were designedand synthesized and the double color nano probes were characterized and identified. This thesismainly divides into three parts, the content is as follows:The first chapter mainly covers the breast cancer and overview of circulating tumorcells, the capture method of circulating tumor cells in vitro and in vivo, the overview ofestrogen receptor and the application of gold and nano gold probe in detection.In the second chapter, in vivo capture and clearance of CTCs based on the transfusion withvein indwelling needle was put forward. Labelled MCF-7cells injected into the vein of wild typemice, rabbits and nude mice could be successfully captured, and2of5labelled MCF-7cellsinjected into the vein of nude mice could be captured. Calculating according to the volume ofblood throughout the body of the experimental nude mice, the minimum capture and detection limit of the in vivo capture method was about2cells/mL blood. This could approach thedetection limit of the best capture and detection method ex vivo (1cell/mL blood). The samplingerror of ex vivo capture method caused by the nonuniform distribution of CTCs could be avoided,the analysis accuracy and sensitivity were improved. CTCs capture was the accompaniment oftransfusion, and there was no extra bodily harm, so would be easily accepted by patients. Thisefficient, simple and less-damage method could be easily popularized with the widespread use ofthe vein indwelling needle.The third chapter, design and synthesis of the estrogen receptor detection double-colornano fluorescent probe was designed and composited. A single fluorescent dye labeled DNAdouble stranded and gold nanoparticles were assembled. Then assembled fluorescence probe isquenched by nanometer gold. Double color fluorescent probes combined the target gene bycomplementary base pairing. The fluorescence chain can be competed and fluorescence of DNAchain can be recovered. Double color probe and complementary base pairing of purpose geneimplemented the specific identification of tumor cells. The method can effectively improve theaccuracy of the specific recognition of cancer.The fourth chapter of this article summarized the circulating tumor cells capture anddetection methods and design and synthesis of double color nano fluorescent probe. Theindwelling needle system operation is simple and convenient, can effectively capture thecirculating tumor cells in the body and clear it out. Double color nanoprobe has highsensitivity, good nuclease stability and biological affinity. It can specificity combined withcirculating tumor cell to obtain more accurate results.
