Ginseng(Panax ginseng C.A.Mey) is one of the precious traditional Chinese medicine in china. Modern medicine study has proved that the main active ingredients in ginseng are ginsenoside and ginseng polysaccharide. Numerous studies show that ginseng can anti-tumor, regulate the central nervous system, improve cardiovascular and hematopoietic system, enhance immunity. Therefore, the establishment of an efficient extraction method is very important and meaningful.The extraction process of ginseng saponin and polysaccharide were studied in this text, the ginseng saponins were also isolated and purified, provide a theoretical basis for the comprehensive development and utilization of ginseng.1)A microwave assisted extraction method for the extraction of ginseng saponins and ginseng polysaccharide have been optimized. Using the single factor experiment and response surface experimental design, the optimum process conditions of microwave assisted extraction of ginsenosides were: liquid/solid ratio of 40:1, extraction time 4 min, extraction temperature 60℃, the ethanol concentration 55%, microwave power 500 W. The average extraction ginsenoside rate was 12.71%, and close to the theoretical maximum yield of 12.75%. By the method of microwave assisted extraction, using water as the extracting agent, the ratio of solid to liquid, the extraction time and the extraction temperature were investigated in the extraction of ginseng polysaccharide, response surface experimental condition has been obtained by optimizing the technology of extraction of polysaccharide in Radix ginseng: liquid/solid ratio 50:1, extraction time 2 min, extraction temperature 60℃. Under the optimal conditions of ginseng polysaccharide extraction achieve 23.32%, is basically the same with response surface theoretical optimum value 24.37%.The method of reflux extraction and microwave assisted extraction were compared in the extraction of ginsenosides and polysaccharide respectively: using 70% ethanol as extracting agent, set the ratio of liquid/solid ratio 20:1, extraction temperature 60℃, bath reflux for 120 min and microwave assisted extraction for 4min, respectively, the extraction rate of ginsenoside respectively were 4.79% and 12.36%; using water as extracting agent, set the ratio of solid to liquid 1:50, extraction temperature 90℃,bath reflux for 120 min and microwave assisted extraction for 2 min, respectively, microwave extraction, ginseng polysaccharide extraction rate were 5.62% and 23.32%.2)Dynamic microwave extraction of ginsenosides has been proposed. the liquid/solid ratio, extraction time, extraction temperature, ethanol concentration, the flow rate of the constant flow pump, ethanol concentration and microwave power were investigated in the extraction of ginseng saponins. the optimum conditions were as follows: liquid-solid ratio of 270:1, microwave time 35 min, microwave temperature 75℃, ethanol concentration of 67%, solvent flow rate of 1.3 m L/min, microwave power of 500 W. 3 parallel experiments weredone under the optimal condition, the average extraction rate of ginsenoside reaches 15.01%, and basically close with the oretical maximum yield of 16.55%. The method of dynamic microwave assisted extraction and microwave assisted extraction were compared in the extraction of ginsenosides, the extraction rate of ginsenoside respectively were 15.01% and 12.71%;3)The four models resin(S-8, AB-8, HPD-100 and polyamide) for separation and purification of the ginsenoside have been compared. HPD-100 was finally chosen as the best one for separation and purification. Resin quality, extraction flow rate, elution volume, elution concentration, extraction time and extraction temperature were investigated. The optimum conditions were as follows: resin quality 2 g, extraction flow rate 1.05 m L/min, elution flow rate of 1 m L/min, elution volume 30 m L, elution concentration 80%, extraction time 40 min and extraction temperature 95℃. The extracting and enrichment of ginsenoside realized at the same time. Dynamic microwave extraction shows better advantage.
