Objective: To investigate the effect of TGF-β1 and Smad3 protein in wound healing process by external application different concentration of Erythropoietin on the acute wound of rats,further investigate the mechanism of EPO promoting wound healing, and the relationship of the EPO and the Smad3 protein.Methods:(1)Divided :72 male SD rats were randomly divided into four group:normal control group n=18,low concentration group n=18, middle concentration group n=18, high concentration group n=18;(2)Moldered: four Group rats were made acute wound model after anesthesia and weighing;(3)Did: cleaned the wound of rats with sterile saline, then wet compress ed the group with 50U/m L EPO, wet compressed the group B with 100U/m L EPO, wet compressed the group with 150U/m L EPO. Nor-mal control group wet compressed with sterile saline. All rats dressed one time a da y, each time 1ml;(4) Recorded: Took a picture everyday. Measure the dia meter by vernier caliper. Calculate the healing rate.(5) Dr-ew materials: k illed the rats, took the skin at the junction of the wound and the normal tissue at the 3,7,14 day.(6) Observed the effect of di-fferent concentration of EPO on the expression of TGFβ1 and smad3 pro-tein in acute wound healing.Results: 1.The seventh day, the use of low concentration and high con-c entration of EPO on rat wound healing rate compared with the control group had statistically significant difference(P < 0.05). The use of m-i ddle concentration of EPO group compared with the control group was s tatistically significant difference(P < 0.001). The third day and the fou-rteenth day were no significant between treatment group and the control group.2. Use of EPO on the wound can promote vessel and fibroblast proliferation in acute wound healing.The effect increases with the increase of concentration.3. Application high and middle concentration of EPO at third day on the wound can promoted expression of TGFβ1.4.Western blot show:The third day, the use of low concentration rat express of smad3 protein was statistically significant difference between control group(P < 0.05).The other groups were no significantly different from control group.Conclusion: 1.Use of EPO in acute wound can improved the wound healing.especially it used until the seventh day.2. Use of EPO promote vessel and fibroblast proliferation in acute wound healing.3. Application high and middle concentration of EPO at third day o n the wound can promoted expression of TGFβ1.4. Application middle concentration of EPO at third day on the wou nd can promoted expression of smad3.5. The mechanism of EPO promoting wound healing is related to the TGFβ1-Smad3 signal pathway.
