Objective To observe the expressiong characteristic of the intracellular transduction medium smad3 and proliferating cell nuclear antigen (PCNA), and research the relationship of smad3 with the chronic ischemic wound and bFGF.Method The study was carried out in three groups: nonischemic group, ischemic group and bFGF group, the model of ischemic ulcer wound on rabbit ear was established in the two later groups. Obtain the wound tissue 3,7,11,15,22 days after the operation,then the general observation was determined to observe the epithelia migration distance and granulation depth, the streptavidin peroxidase(SP) immunohistochemistry methods was used to detect the protein expression of smad3 andPCNA.Result There are significant difference among the healing speed of the three groups, the nonischemic group is faster than the others. At the same time, the expression rate of smad3 and PCNA of nomschemic group is more higher than the other two groups. bFGF can increase the expression of PCNA in ischemic wound, but not smad3.Conclusion smad3 takes part in the process of inflammation and granulation formation. The low expression of smad3 and PCNA may participate in the happening of the chronic wound. bFGF can increase the number of cell that is breeding,but can not impact the expression of smad3 in the ischemic wound. Smad3 .which is one of the intracellular signaling transduction medium may not take part in the intracellular signaling transduction of bFGF.
