Objective:Establish a stable and effective model of insulin resistance,to observe the effects of active component of Radix Isatidis (ACRI) on Insulin resistance cells and animals, At the same time, we investigated its possible mechanism by measuring the related Factor and protein expression expression.Methods:Vitro 3T3-L1 preadipocytes,and use the cocktail method to induced 3T3-L1,Oil red O staining was carried out for investigating the effect of 3T3-L1 induced, High glucose and insulin induced the cell become insulin resistance model, Glucose oxidase and MTT (MTT) method to determine the success and stability of the model, ELISA was used to detect the influence of ACIR on TNF-α,then we used western blot to detect the expression of PI-3K p85,clut4 protein; we rendered mice insulin resistant and diabetic by feeding high glucose and fat diet together with peritoneal injection of STZ and then treated it with different ACIR dose groups, comparing to RosiglitazoneMaleate.then we observed the effects of the drugs on mice’s body weight, food intake, fat weight,liver weight, pancreas weight and the general state of the animal, and detected serum total cholesterol (TC), triglyceride (TG), low density lipoprotein (LDL), high density lipoprotein (HDL) glucose (GLC),Free fatty acid (FFA), fasting insulin (FINS), fasting blood glucose concentration (FBG), serum superoxide dismutase (SOD) and malondialdehyde (MDA),at last,we used HE staining and immunohistochemical staining to observe the effects of ACIR on the liver and pancreas.Results:High glucose with insulin can successfully induce insulin resistance cell model, and can stability in 48 hours,ACRI within 25~100μg·mL-1 concentration range can reduce cell supernatant glucose and TNF-a levels, increase the expression of cell PI-3K P85, clut4 protein. In vivo tests,we had successfully replicated the type 2 diabetic rat model, ACRI can significantly improve the symptoms of diabetic rats, delay the mitigation weight of rats and reduce rats liver weight and liver index. ACIR can decrease serum TC, TG, LDL,FFA,FBG,FINS,MDA value and increase serum HDL, SOD, ISI,Under the action of ACIR,glucose tolerance in the rats has been improved, liver and pancreas damage has been some restoration.Conclusion:(1)ACRI has the effect of improving insulin resistance cells;(2)ACRI improve the role of insulin resistance may be through decrease supernatant glucose and TNF-a levels, enhancing the expression of PI-3K p85, CLUT4 protein;(3)ACRI can improve the insulin resistance、glucose metabolism and lipid metabolism in type 2 diabetes rats, furthermore, it has the effect to protect the liver and pancreas.
