Objective: Our objective was to Research Wen Tong method on the anti-tumor function of the mice which were transplanted with S180 sarcoma cells and its relationship with adjust a tumor-burdened mice immune function,and to investigate the immunological mechanism of the anti-tumor effect.Method: 72 healthy Kunming SPF mice, both male and female mice randomized,marked as blank control group, a tumor-burdened in the control group; low dose group,the middle dose group, high dose group of the Chinese medicine(To fill the stomach through the mouth with the Chinese medicine of Wen Tong method, the low dose group take the equivalent of traditional Chinese medicine raw medicinal materials of 1.30 g/ml of liquids, the middle dose group take the equivalent of TCM raw medicinal materials of 2.56 g/ml of liquids, and the dose of the high amount group is equivalent to TCM raw medicinal materials of 3.83 g/ml solution),Cytoxan injection intraperitoneal injection(CTX) group.There were six groups, and each group contained12 mice.Blank control group and the tumor-burdened group, were given gavage with normal saline in the next day, 0.2ml/start, only 1 times a day, after builded models successfully;we drench the mice with Chinese herbs in high、middle、low dose groups in the next day with the corresponding concentration of traditional Chinese medicine decoction lavage, 0.2 ml/start, only 1 times a day;according to the numbers increase or decrease in weight to adjust the dose after a week.CTX group was given CTX intraperitoneal injection of 30mg/kg(2mg/ml) after 72 ours, every other day for 1times each.All groups of experimental mice were stopped on the twelfth day after builded models. Determinated the inhibition rate(IR)of drugs on each tumor-burdened mice and between groups of thymus index changed; Modified tetramethyl azo thiazole blue(determined by MTT colorimetric method to determine the natural killer cells(NKcell) killing activity; Using flow cytometry instrument(FACS) method to determinate the CD4+T lymphocytes in peripheral blood of mice, CD8+ cells andCD4+/CD8+ ratio changes; Application of double antibody enzyme-linked immunosorbent analysis method(ABC-ELISA) of lotus S180 sarcoma in mice to observe Wen Tong method how to influence the interferon-γ(IFN-γ), interleukin-2(IL-2)levels in the peripheral blood.Results:( 1) In the Tumors quality and inhibitory rate experiment, compared with the tumor-burdened control group,the low, middle and high dose of TCM groups and the CTX group,the differences were statistically significant(P<0.05). Compared by CTX group,the tumors quality of the low and high dose of TCM groups,the differences were statistically significant(P<0.05).(2) Compared with the blank control group, the thymus index of the CTX group,the tumor-burdened group,the low, middle and high dose of TCM groups,the differences were statistically significant(P<0.05).Compared with the tumor-burdened group,the thymus index of high, middle and low dose of TCM groups were significantly increased,the differences were statistically significant(P<0.05).(3) Natural killer cells killing activity experiment showed that middle dose of TCM group increased the effect of killing activity of NK cells obviously,compared with the low and high dose of TCM groups,the differences were statistically significant(P<0.05).Compared with the CTX group,killing activity of NK cells of the middle dose of TCM group, the difference was statistically significant(P<0.05).(4) Compared with the blank control group,tumor-burdened group, low and high dose of TCM groups, and CTX group, CD4+/CD8+ ratio was obviously in a state of imbalance,the differences were statistically significant(P<0.05).Wen Tong method can change the number of CD4+ and CD8+ cells in peripheral blood and significantly increased the ratio of CD4+/CD8+ cells.(5) Compared with tumor-burdened control group, the IFN-γ level in peripheral blood of low, middle and high dose of TCM groups, were significantly elevated, the differences were statistically significant(P<0.05),and the IFN-γ level of CTX group was significantly lower than the tumor-burdened group(P<0.05).Compared with tumor-burdened control group, IL-2 level in peripheral blood of middle and high dose of TCM groups, was significantly elevated,the differences were statistically significant(P<0.05).and we found that Wen Tong method could make the IFN-γ, IL-2increased, so as to play its anti-tumor effect in immunology.Conclusion:( 1) Wen Tong method can improve the thymus index of a tumor-burdened mice,to a certain extent,can enhance the immune function of central immune organs-the thymus,Wen Tong method of traditional Chinese medicine.( 2) Wen Tong method may be by raising a tumor-burdened mice spleen NK cell killing activity,to exert its function of killing tumor cells.(3)Wen Tong method may be by raising CD4+ cells, reduce the CD8+ cells, and significantly higher CD4+ / CD8+ cells subgroup ratio, thus play a role in regulating cellular immune function.(4)Wen Tong method cytokines may be by raising the secretion of IL-2, IFN-γ, so as to exert the function of inhibiting tumor cell proliferation.
