Objective: Based on Fas apoptosis pathway, taking the effects of LIUWEI DIHUANG decocting-free granule on pregnancy result of the Kidney-YIN deficiency type patient, who is carried out in vitro fertilization and embryo transplantation(IVF-ET), as an entry point, discuss the “ key gene- apoptosis pathway- key protein” network regulation of LIUWEI DIHUANG decocting-free granule on apoptosis Signaling Pathway, and indicate the scientific connotation of “kidney-nourishing and YIN-promoting propagation” theory substantially.Methods: Choose 66 cases who are discriminated as YIN-deficiency type of the kindey IVF-ET patients from clinic, divide into treatment group 33 cases(LIUWEI DIHUANG decocting-free granule, gonadotropin-releasing hormone releasing agent(GnRH-a)and gonadotropin(Gn)) and placebo group 33 cases(placebo decocting-free granule, GnRH-a and Gn) according to random number table method. Patients of treatment group take LIUWEI DIHUANG decocting-free granule for 2 menstrual cycles, and patients of placebo group take placebo granule for 2 menstrual cycles. Carry out standard long therapy ovulation-inducing treatment after above treatment. Gather the follicular fluid of both groups separately, extract granular cells, test the expression level of key protein Fas and Caspase-3 in Fas mediated apoptosis pathway by using western-blot. Compare the difference changes of deficiency of the Kidney-YIN symptom score, the expression of key protein Fas and Caspase-3, GnRH-a dosage, Gn dosage and number of days, number of gained eggs, high-qualified eggs rate, high-qualified embryos rate and clinical pregnancy rate before and after taking drug.Results: 1. Compared to placebo group, the expression of key protein Fas and Caspase-3 in FAS mediated apoptosis signaling pathway of LIUWEI DIHUANG decocting-free granule treatment group reduces significantly, and the difference between two groups is statistically significant(P<0.05); 2. Compared to placebo group, Gn dosage and number of days of LIUWEI DIHUANG decocting-free granule treatment group is less than placebo group, and the difference between two groups is statistically significant(P<0.05); The difference of GnRH-a dosage and number of gained eggs between two groups is not statistically significant(P>0.05); 3. Deficiency of the kidney YIN symptom of LIUWEI DIHUANG depicting-free granule treatment group improves significantly after treatment, and the Gn dosage and number of days, high-qualified eggs and high-qualified embryos rate are all higher than placebo group(P<0.05), the difference is statistically significant(P<0.05).Conclusions: 1. LIUWEI DIHUANG decocting-free granule can reduce the expression of key protein Fas and Caspase-3 in Fas mediated granular cells apoptosis signaling pathway, inhibit Fas mediated granular cells apoptosis signaling pathway, reduce granular cells apoptosis, improve ovarian "storage" function, improve the quality of egg cell, and increase clinical pregnancy rate of IVF-ET patients as a result. 2. LIUWEI DIHUANG decocting-free granule can reduce the expression of key protein Fas and Caspase-3, and the expression of key protein Fas and Caspase-3 can be used as important index to forecast egg development potential and IVF-ET pregnancy result. 3. Based on Fas mediated granular cells apoptosis signaling pathway, indicate the scientific connotation of “kidney-nourishing and YIN-promoting propagation” theory from the point of the “ key gene- apoptosis pathway- key protein”.
