Objective:To investigate the expression of transcription factor Ets-1, Runx3 in patients with psoriasis of blood heat syndrome in peripheral blood and the prescription of damp heat clear changes in the expression of Ets-1, Runx3 before and after treatment, further explains the transcription factor Ets-1 and Runx3 in patients with vulgaris psoriasis of blood heat syndrome pathogenesis of damp heat clearing mechanism, reveals the micro mechanism of treating blood heat syndrome of vulgaris psoriasis.Method:select July 1,2013 September 1,2014 Hospital Affiliated to Shandong University of Traditional Chinese Medicine Department of dermatology in out-patient and ward patients with vulgaris psoriasis of blood heat syndrome 28 cases set to treatment group, select the Shandong University of Traditional Chinese Medicine hospital health examination center in 20 healthy people who set up as the healthy control group.The treatment group was given oral prescription SRQ and modified, one dose daily,2 month observation period. Extraction after treatment in the treatment group and the healthy control group in the early morning fasting peripheral blood 4ml, total RNA was extracted from the peripheral blood of TRIzol method; detection of samples of RNA concentration, RNA transcription by TaKaRa kit; design reference gene, using real-time quantitative PCR (Real-timePCR) method was used to detect the target gene and reference gene CT expression of each objective value; calculation gene dosage by 2-Delta CT method; the data were analyzed using SPSS 17.0 software.Results:the quantity is obviously higher than that in healthy control group Ets-1mRNA expression of treatment group before treatment in peripheral blood, the difference was statistically significant (t=2.91, P=0.08<0.01); the expression of Runx3mRNA was slightly lower than that in healthy control group, no significant difference (t=-0.07, P=0.994>0.05). The treatment group were given modified prescriptions of treating damp heat after peripheral blood Ets-1mRNA expression significantly decreased, the difference was statistically significant (t=2.817, P=0.01<0.05); the expression of Runx3mRNA was slightly increased, no significant difference (t=-1.125, P=0.273>0.05). Is slightly higher than that of the healthy control group the treatment group after treatment the expression of Ets-lmRNA in peripheral blood, but the difference was not statistically significant (t=0.422, P=0.676>0.05); the expression of Runx3mRNA was slightly higher than that in healthy control group, but the difference was not statistically significant (t=0.611, P=0.546>0.05).Conclusion:the abnormal expression of Ets-1 and Runx3 in patients with vulgaris psoriasis of blood heat syndrome of damp heat in the peripheral blood; prescription Qing Jiawei may regulate the transcription factor Ets-1 and Runx3 expression in indirect effects on T lymphocyte differentiation, can play a role in the treatment of patients with vulgaris psoriasis of blood heat syndrome. |
