Objective:To explore the establishment of iodine deficiencygoiter ani mal model;To investigate the effects of huoxuexiaoying tablet for thyr oid function and weight in Iodine deficiency goiter of rats;To detect s everal cytokines related levels of Rats;To investigate the mechanism of pathogenesis and therapy on huoxuexiaoying Tablet of goiter from angi ogenes is and apoptosis angle; Provide relevant experimental data con firm theexact effect of huoxuexiaoying Tablet in the treatment of goiter,to further promote its application in clinical practice. Methods:1.To prepare the model of iodine deficiency goiter in rats:50 F emale Wistar rats and 50 Male Wistar rats,Four weeks,Weighs about 90‐120g,Divided 5 groups with Wistar masculinity rats at random:the Normal control group,the model control group, the huoxuexiaoying tablet group, the Euthyrox group and the huoxuexiaoying table Comb ine Euthyrox group(Combined group),and 20 rats every group. Normal group of rats fed with normal diet and double distilled wate feeding,The remaining four groups of rats are used by iodine‐deficient areas of crops produced processed formulated into a low iodine diet w ere fed, and deionized water for drinking.Feeding three months later, t he use of thyroid palpation, combined with thyroid ultrasound to deter mine the successful model.2.Administration:Continue normal rats fed a normal diet anddouble distilled water,The rest of the rats continue to be low iodine diet an deionized water feeding,Normal group and model group as a control does not give medication.the huoxuexiaoying tablet group were treate with huoxuexiaoying powder at 10mg/g.d orally, the L-T4 group were given with 0.2ug/kg.d L-T4 by intragastric(i.g.).The Combined group were given with huoxuexiaoying powder at 5mg/g.d orally and 0.1ug/kg.d Euthyrox by i.g. administration;A total of three months of treatment3. Specimen collection and data recording:2% lidocaine, according to 0.2ml/100 g for 5 groups of rats were anesthetized by intr aperitoneal,Abdominal aortic blood,Each rat was collected four each, e ach tube 3‐5ml, and well marked,Separation of serum,Placed in ‐20℃ r efrigerator for up to detect.Blunt dissectionrat thyroid tissue,Precision w eighing,The rats were sacrificed humanity.4.Radioimmunoassay detect three free triiodothyronine(FT3), free t hyroxine(FT4), promote thyroid hormone(TSH).5.ELISA method :to detect density of VEGF in serous of rats6.ELISA method :to detect density of sFas、sFasL in serous of rats7.OUTCOME MEASURES:Differences between treatment and control groups after comparing treatment of Thyroid Weight,Thyroid function an d the levels of VEGF、sFas、sFasL Results:1.The results indicate thyroid tissue weight,Normal control gr oup, a significant increase in thyroid weight of the model control grou p of rats(P<0.01), and thus illustrate the method by controlling rats d aily intake of iodine to prepar iodine deficiency goitre rat model is fea sible, Preparation ofthe success of this animal model; L-T4 thyroid weig ht group and model group comparisons and promoting blood Xiaoying piece group though has declined, but there was no statistically significant; comparison with the model group, a joint group of thyroidweight d ecreased significantly(P<0.05), the difference was statistically significant.Combined with the blood Xiaoying description sheet and L-T4 treatme nt of iodine deficiency goiter isbetter than the two drugs alone.2.The results of thyroid function: Compared with normal group, m odel group FT3, FT4 decreased, TSH increased(P<0.01), showing the sta te of hypothyroidism; compared with the model group, blood group Xi aoying piece of thyroid function did not change significantly, no statisti cally significant; compared with the model group, L-T4 group and the c ombination group FT3, FT4 increased, TSH decreased significantly(P<0.05), there are statistically significant.3. Detection of serum VEGF concentration showed that: Compared with the normal group, significantly increased serum concentrations of VEGF in model group(P<0.01); compared with the model group, VEGF serum concentrations were decreased in rats treated groups(P<0.01); compared with euthyrox group, blood Xiaoying piece group and the co mbination group rat serum VEGF concentration was no significant differ ence(P>0.05). The results showed that Euthyrox and promoting blood Xiaoying pieces are cutby a modest serum VEGF levels and the decreas e of combination therapy is more obvious.4.The serum s Fas, s FasL concentration test results show: compared with the model group decreased serum s Fas concentration combined group of rats was significantly(P<0.01), while Euthyrox group and pro moting blood Xiaoying piece group declined, but there was no signifi cant difference(P>0.05); compared with the model group, combination therapy significantly increased serum concentrations of s FasL rats(P<0.01), which is higher than euthyrox group and activating blood infla mmatory tablets group(P<0.05), also higher than the normal group(P<0.05), compared with the model group, euthyrox group and promoting blood serum sFasL concentration Xiaoying always make rats piece grou p increased(P<0.05). Conclusion:1. huoxuexiaoying tablet of anti‐goiter role in promoting blo od circulation and small Xiaoying side effects forthyroid function, fewer side effects, better safety performance.2. vascular endothelial growth factor(VEGF) in the pathogenesis and treatment of goiter plays a significant outcome, while huoxuexiao ying tablet of thyroid tissue hypertrophy inhibition mechanism may be through reduction of serum VEGF concentration to achieve.3. apoptotic factors(sFas, s FasL) and goiter incidence and treatme nt are closely linked, bhuoxuexiaoying tablet may moderate reduction o f serum sFas concentration, and a modest rise inthe concentration of s erum sFasL induce apoptosis active thyroid, thus even narrow purpose of suppressing thyroid enlargement of the thyroid tissue.
