Objective: To make Chrysanthemum stems and leaves(CSL) used as natural antioxidant,and study on the preparation process.Methods: A quality control system of CLS was built up. HPLC-MS technique was applied in the chemical identification. By analyze the content of the total flavonoids and chlorogenic acid to optimal the parameters in the extraction and the purification process. In-vitro antioxidant capabilities of the intermediate was determined by the DPPH assay and the FRAP assay.Results: A quality control system of CSL was built up,the content of impurity should limited to 4%, and the moisture content should limited to 10%, the ethanol-soluble extractive should limited to 22%, the content of total flavonoids and chlorogenic acid should limited to 1.5% and 0.05%. 10 kinds of chemical composition was identified in the HPLC-MS. The optimal extracting process was established as follows: the CLS was extracted with 50% ethanol(1:20,g/m L) under reflux for 2 h and repeated three times. AB-8 macroporous resin was selected to be the most suitable resin for the purification. The final purification process was as follows: the concentration of the extraction solution was 0.5 g/m L. Then the column was eluted by water and 70% ethanol in the amount of 3 BV respectively. The antioxidant capabilities of the intermediate was stronger than CSL significantly, while little weaker than ascorbic acid.Conclusion: The preparation process of the intermediate from Chrysanthemum leaves and stems was steady, can be used as natural antioxidant.
