To make a summary of therapeutic principle and method about Chinese medicine curing liver damage by literature review, and to analyze the drug using regularity and research progress through summarizing the clinical and experimental study about liver damage. Cornus was the research object on the base of previous study, and D-Ga1N/TNF-α was the L-02 cell damage model to prove the effect of Cornus on it.Theoretical studyWith "liver damage", "liver disease", "liver" as keywords to search in China National Knowledge Internet, we summarize the therapeutic principle and method about Chinese medicine curing liver damage, and classify the related compound and single medicine research literature at the same time. Then class statistics all the related Chinese medicine according to the Chinese medicine teaching material and analyze the drug using regularity and research progress. Exogenous damp and hot, improper diet, and drug poisoning were liver damage’s pathogenesis. Liver dysfunction, dysfunction of the spleen in transport, and retention of damp-heat, and blood deficiency for stagnation of the circulation of vital energy were onset and process of an illness pathogenesis of liver damage. It would result in hypochondriac pain, jaundice, tympanites, and accumulation. The disease location was liver, and spleen, and stomach were also damaged, and with days went on, kidney was damaged too.The compound research literature results showed that soothing the liver, tonifying the spleen, harmonizing the stomach were the main curing regularity; the Chinese herbal compound results showed that antipyretic, tonic drugs and blood-quickening drugs were the most used on curing liver damage. The single Chinese herbal statistical results showed the same result with compound results. Meridian tropism statistical results showed that Chinese herb on curing liver damage were mainly into liver, and then spleen and stomach.Cornus was commonly used as a tonic kidney drug. Its chemical composition, pharmacologic action and the progress of curing liver damage were summarized through literature review.Experiment researchPreparing CIG, cultivating human hepatocytes(L-02) in vitro, making hepatic cell lesion model by D-Ga1N/TNF-α were the first step of the experiment. Control group,model group, CIG high dose group, medium dose group and low dose group were set up. After the cell processing, the percentage of cell viability was evaluated by MTT method, and the total antioxidant capacity(T-AOC), the content of superoxide dismutase(SOD), malondialdehyde(MDA), lactic dehydrogenase(LDH), glutathione peroxidase(GSH-Px) were used indicators to show the antioxidant ability of injured hepatic cell. The cell metamorphosis of each group was observed by Hochest-33258 staining. The apoptosis rate of each group was tested by flow cytometry. The expression levels of phosphorylase protein kinase ER like kinase(p-PERK), phosphorylase eukaryotic translation initiation factor 2(p-Eif2 a), activating transcription factor-4(ATF-4), CCAAT/enhancer-binding protein-homologous protein(CHOP), and apoptosis associated proteins cysteine aspartyl protease-3(Caspase-3) were tested by western-blot analysis.Results: The MTT results showed that the apoptosis rate was 38% by 44μg/ml D-Ga1N and 100ng/mlTNF-α injured for 12 hours, and CIG high dose group, medium dose group and low dose group improved the injured cell’s inhibition rate by 21%,13%,8% respectly, the difference was significant comparing the model group(P<0.05). The content of SOD was improved significantly, and the content of LDH was reduced significantly in CIG high dose group, medium dose group and low dose group(P<0.05). The content of MD was reduced significantly in CIG high dose group, medium dose group(P<0.05), but in the low dose group, the diference was not significant. The cell metamorphosis of CIG high dose group, medium dose group and low dose group were better than model group, CIG could protect the integrity of the injured cell’s cell membrane. The flow cytometry results showed that the apoptosis rate of CIG high dose group, medium dose group and low dose group were reduced than model group significantly(P<0.05). The activity of T-AOC and GSH-Px were improved significantly in CIG high dose group, medium dose group and low dose group(P<0.05), comparing with model group. The expression levels of p-PERK, (p-Eif2a, ATF-4, CHOP, and Caspase-3 were reduced in CIG high dose group, medium dose group and low dose group.Conclusion:CIG could protect L-02 injured by D-Ga1N/ TNF-α, and improving the antioxidant ability of injured hepatic cell and reducing the expression levels of related protein in endoplasmic reticulum stress(ERS) apotosis pathway were the possible mechanism.
