Objective:To detect the expression of Oct-4 protein in chronic cervicitis,cervical intraepithelial neoplasia(CIN)and cervical cancer tissue by using immunohistochemical technique,and to explore its clinical significance and relationship with HR-HPV infection in the occurrence and development of cervical cancer,hope to provide a theoretical basis for early correct diagnosis,risk assessment,effective treatment and follow-up monitoring of it.Methods:To detect the expression of Oct-4 protein in 44 cases of cervical cancer,43 cases of CIN,15 cases of chronic cervicitis tissue by using immunohistochemical technique,and to analyze its relationship with patient age,clinical stage,histological type,degree of differentiation,lymph node metastasis,correlation with HR-HPV and multiple HR-HPV infection in cervical cancer.Results:1.The rate of Oct-4 protein expression in chronic cervicitis,CIN and cervical cancer tissue was respectively 20.0%(3/15)、27.9%(12/43)、79.5%(35/44),the difference was statistically significant(P<0.05).Difference between chronic cervicitis and CIN was no statistically significant(P>0.05).Difference between chronic cervicitis and cervical cancer was statistically significant(P<0.05).Difference between CIN and cervical cancer was statistically significant(P<0.05).2.Oct-4 expression in cervical cancer has no relationship with patient age,clinical stage,histological type,lymph node metastasis(P>0.05),but negatively correlated with degree of differentiation(P<0.05).3.The rate of Oct-4 protein expression in chronic cervicitis,CIN and cervical cancer tissue with HR-HPV infection was respectively 33.3%(2/6) 、 31.3%(10/32) 、83.3%(25/30), the difference was statistically significant(P<0.05).Difference between chronic cervicitis and CIN was no statistically significant(P>0.05).Difference between chronic cervicitis and cervical cancer was no statistically significant(P>0.05).Difference between CIN and cervical cancer was statistically significant(P<0.05).4.The rate of Oct-4 protein expression in chronic cervicitis,CIN and cervical cancer tissue without HR-HPV infection was respectively 11.1%(1/9) 、 18.2%(2/11) 、71.4%(10/14),the difference was statistically significant(P<0.05).Difference between chronic cervicitis and CIN was no statistically significant(P>0.05).Difference between chronic cervicitis and cervical cancer was statistically significant(P<0.05).Difference between CIN and cervical cancer was statistic-ally significant(P<0.05).5.The rate of Oct-4 protein expression in chronic cervicitis,CIN and cervical cancer tissue with single HR-HPV infection was respectively 25.0%(1/4)、27.8%(5/18)、84.6%(22/26),the difference was statistically significant(P<0.05).Difference between chronic cervicitis and CIN was no statistically significant(P>0.05).Difference between chronic cervicitis and cervical cancer was no statistically significant(P>0.05).Difference between CIN and cervical cancer was statistically significant(P<0.05).6.The rate of Oct-4 protein expre-ssion in chronic cervicitis,CIN and cervical cancer tissue with multiple HR-HPV infection was respectively 50.0%(1/2) 、35.7%(5/14)、5.0%(3/4),the difference was no statistically significant(P>0.05).7.Oct-4 expression in cervical cancer has no correlation with HR-HPV and multiple HR-HPV infection(P>0.05).Conclusion:Oct-4 expression in cervical cancer is significantly higher than CIN whether there is a HR-HPV infection or not. Oct-4 expression in cervical cancer has no relationship with patient age,clinical stage,histological type,lymph node metastasis,but negatively correlates with degree of differentiation.What is more,it has no correlation with HR-HPV and multiple HR-HPV infection.It indicates that Oct-4 playes a main role in the occurrence and development of cervical cancer,HR-HPV infection just playes a catalytic role in the process.Detection of Oct-4 expression maybe helpful for early diagnosis,risk prediction,targeting therapy of cervical cancer.All those still need to be explored.
