Heat stroke is a kind of sepsis like syndrome caused by high temperature, hence resulting in a series of multiple organ dysfunction syndromes(Multiple Organ Disfunction Syndrome,MODS).Under the special, mal-conditionor trauma situation, lung is one of the most vulnerable organs to be damaged. Occurring of heatstroke is often associated with lung functional damage; however, there is lack of investigations regarding heatstroke related lung injury induced by hot desert environment in rats. Curcumin has extensive pharmacological effects, e.g., effects of anti-oxidation, anti-inflammation, anti-tumor. We hypothesize that curcumin may have function of viscera protection on rats after heat stroked under the condition of hot desert environment. In this study the protective effect of different concentration of curcumin on rats, heat stroke was investigated by using rat heat stroke model induced by hot desert environment. We aim(1)To study establish a hot desert environment induced rat heatstroke not an aesthetic model, and through the rat in all kinds of physiological indexes, active in hot desert environments, lung wet dry weight ratio, lung pathological changes and ultrastructure changes and alveolar lavage fluid analysis, explore the hot desert environment change characteristics of different damage degree of rat lung.(2) by curcumin preventive medication, to explore whether curcumin on the hot desert environment induced lung injury plays a protective role in rats. Methods of experiment: The 120 selected 6-8week-old male SPF grade SD rats were randomly divided into control group, low-dose curcumin pretreated group, middle-dose curcumin pretreated group, high-dose curcumin pretreated group and solvent group. The control groups were fed as normal, the three curcumin pretreated group were gavaged with different dose of curcumin for 7 consecutive days, the solvent group were gavaged equal volume of solvent for 7 consecutive days. Then all the rats were put into the dry heat environment(temperature of 41 ℃, humidity10%,ultraviolet radiation 40W/m2) in The Simulated Climate Cabin for Special Environment of Northwest of China, The vital signs of the rats were observed, The core body temperature of the rats were monitored for every half an hour, the dead time were recorded. The lung histopathology and ultrastructure change of observation, lung wet dry weight ratio and lung lavage protein detection analysis. Results show(1) of curcumin three dose groups after the treatment, the core body temperature is lower than the control group and solvent group.(2) the low, middle and high dose group into the tank after 90 min, 180 min, 240 min three time points lung wet dry weight ratio(W/D) were higher than in the control group and solvent group; The control group, the solvent with low, medium and high dose group of lung wet dry weight ratio(W/D) there is significant difference(P < 0.05);Low, medium and high dose of lung wet dry weight ratio between the three way was no significant difference(P < 0.05).(4)into the tank after 90 min time points, 180 points and 240 min time point in time, control group, lung tissue pathology score in solvent group were slightly higher than in low, middle and high dose group(P < 0.05);The degree of lung injury of lung tissue were lighter than control group and solvent.(5) in the control group and solvent group lung lavage fluid of thetotal amount of protein at three time points were higher than in low, middle and high dose group, low, medium and high dose is at all-time points in the group and the difference was not significant(P > 0.05). We drew the following conclusion(1) we have successfully established the rat without anesthesia heatstroke modal of dry-heat environment of desert for the first time.In order to further explore the hot desert environment heatstroke areas laid a solid foundation.(2) The hot desert environment heat stress can cause lung injury in rats, and increased gradually along with the extension of heat stress time; prompt heat treatment in a hot desert environment should be paid attention to strengthen prevention and protection of the lungs.(3)Of curcumin can reduce rats in the hot desert environment caused by pulmonary edema and lung tissue damage, has some lung protective effects. Early for curcumin can extend the hot desert environment heatstroke rat survival time, and effective protection for their lungs.
