[Purpose] The thyroid is the largest endocrine gland of human and animals, which plays a very important role on the growth and metabolism. Some enverinmental chemicals had a significant impact on the thyroid gland, which can interfere with thyroid function, induce autoimmune thyroid diseases and cause thyroid tumors. Pesticieds are the largest part of thyroid disrupting chemicals. As a non-selective herbicide, Amitrole showed the abilityof interfere with the synthesis of thyroid hormones, lead to thyroid cancer and thyroid hyperplasia, but the mechanism is not clear. In this study, human thyroid cells Nthy-ori-3-1 were used to research the changes of thyroid-related genes under exclude cytotoxic effect level of Amitrole.[Method] MTT method was applied to study the effect of amitrole on the proliferation of Nthy-ori-3-1 cells. The cells were treated with 0 μg/ml,1 μg/ml,10 μg/ml,100μg/ml dose of amitrole in 24 hours. Single standard Agilent expression profiling microarray was used to screen the differentially expressed gene of the cells. Five genes were seleced to verify the results by Real time quantitative PCR technique.[Result] MTT assay showed that, Amitrole has no significant change on proliferation of cells within the dose of 1~100μg/ml. There are 90 genes showed differences in microarray, including 55 up-regulated genes and 35 down-regulated genes. The qPCR results were consistent with the microarray. AGT, CA11 are up-regulated genes and MUC12, RAPH1, POLR1Aare down-regulated genes.[Conclusion] The influence of amitrole on the cells independent of cytotoxic at the dose of 1~100μg/ml. In the difference expression genes, AGT, CA11, MUC12 plays an important role in the development of thyroid tumors; RAPH1, POLR1A can impact the cell structure, including protein synthesis, modification, metabolism and Cytoskeleton assembly.
