The Study Of Cytotoxicity Of CIK Cells Against Tumor Cell In A Length Of Time

CIK cells are a set of T cells, which can proliferate abundantly in vitro. CIK cells can kill various kinds of tumor cells. Furthermore, there are few adverse effects by the treatment with CIK cells. All of those promote the widely using of CIK cells in the clinical treatments. However, the current assays only present the results of cytotoxicity of CIK cells against tumor in a short time. It is unclear that after a length of time of interaction between CIK cells and tumor cells, the CIK cells still have cytotoxicity against tumor.In this research, we observed the major subsets of CIK cells during the culture. And a new method was established, which is used to study the substance cytotoxicity of CIK cells. Co-culturing CIK cells with breast tumor cell line SBKR3, at most for96h. We observed the behaviors of them and the interaction between them.The results showed that CIK cells are mainly CD3+T cells that reach97.67±1.24%in the day12; the major effector cells, CD3+CD56+cells and CD3+CD8+CTL, are gradually increased during the induction in vitro. CD3+CD8+CTL cell reach a plateau (86.43±7.17%) around d16days. The NKG2D+cells also increasing during the culture and reach the plateau (90.3±7.14%) around d16. The CD4+CD25high regulatory T lymphocytes is only3.03±2.03%in the day16. Then, according to our results, it indicates that the CIK cell can kill tumor cells in a length time. In reality, at least we observed that its cytotoxicity lasted96hours. Meanwhile, there are almost no differences among the cytotoxicity of the CIK cells cultured for12to20days. The ratio of target cell number to the effect cells was taken as1:20. And there is no obvious impact observed on the proliferation of CIK cells by SKBR3. Summing up the above points considered that the d14-16is the best harvest time.The cytotoxicity of CIK cells to tumor lasts a long period of time. As a new adjuvant therapy, CIK treatments show effectiveness and potential in clinical treatments.
