Kunming 650031, ChinaObjective:To explore the effect of aromatase inhibitors on the transgenic salivary adenoid cystic carcinoma (ACC-83) so as to provide a scientific theoretical basis for clinical endocrine therapy of salivary gland cancer.Materials and methods:The apoptosis of ACC-83 and was detected using MTT and flow cytometry (FMC) technology, respectively. The expression of aromatase in ACC-83, ACC-LM, ACC-2 and transgenic ACC-83-CYP19 were assessed using immunohistochemistry and western blot technology, respectively.Results:1.The expression of aromatase was detected in the cytoplasm of ACC-2, ACC-LM and ACC-83.2.The expression of aromatase P450 was higher in ACC-83-CYP19 than in ACC-83,3.Different concentrations of letrozole were used to construct ACC-83 and ACC-83-CYP19, including 25nmol/L,50nmol/L,100nmol/L and 200nmol/L.The inhibition rates are 62.93%,74.82%,82.50%,85.51% in ACC-83-CYP19 and the inhibition rates are 65.27%,69.71%,70.59% and 67.43% in ACC-83 by MTT.4. Letrozole can induce the apoptosis of ACC-83 and ACC-83-CYP19. The apoptosis rate increased following the increasement of drug concentration.5. Letrozole could reduce the expression of aromatase in ACC-83 and ACC-83-CYP19 cells.Conclusions:1.Aromatase was expressed in adenoid cystic carcinoma cell line ACC-83, ACC-LM and ACC-2.Adenoid cystic carcinoma may be associated with abnormal expression of aromatase.2. Letrozole can inhibit the growth of cell and induce apoptosis in ACC-83 and ACC-83-CYP19 cells.
