Effects Of Letrozole On Expressions Of Bcl-2and Bax In Endometriotic Rats

ObjectiveTo establish an endometriotic rats.To determine the treating effect of aromatase inhibitor by measuring the ectopic lesions volume before and after the treatment, the expression of B cell lymphoma/lewkmia-2(bcl-2) and bax in the ectopic lesions.Methods40female unpregnant Sprague-Dawley(SD) rats were randomly selected to construct endometriosis model in rats’abdominal wall by autotransplantation in estrus.4weeks after surgery (before the treatment), the shape and size of the SD rats’ectopic lesions were observed and measured.30induced successfully SD rats model were randomly divided into2groups(n=15):Letrozole group:a dose of0.26mg/Kg letrozole was given orally every day for3weeks;Model group as control:the same volume of saline (NS) was given orally every day for3weeks.7weeks after surgery(3weeks after the treatment), SD rats were killed to be observed and measured the shape and the size for the second time. The ectopic lesions were cut off to do HE tests. Expression level of bcl-2and bax were assesed by immunohistochemistry and reverse transcription-polymerase chain raction(RT-PCR).Results1.30SD rats among all of the40ones formed endometriotic lesions,the suecessful ratio was75.0%(30/40).2.Before the treatment,no difference was existed in the size of the ectopic lesions between the both groups (P>0.05);3weeks after the treatment,the size of the letrozole group reduced markedly compared with the previous administration(P<0.05); The size of the model group as control had no increase compared with the previous administration(P>0.05).3.After the treatment, the ectopic endometrial glands were withered,the blood vessel were reduced in the letrozole group,while the glands were inereased, the blood vessel were rich in the model group as control.4.3weeks after the treatment,the protein and mRNA of bcl-2and bax were both detected in the ectopic lesions.Compared with the model group as control,the level of bcl-2was lower, while the level of bax was higher,so the rate of bax/bcl-2was higher in the letrozole group (P<0.05).Conclusionl.Autologous transplantation of endometria is with a high achievement ratio.2.Aromatase inhibitors have the ability to treat endometritic rats by suppressing endometrial epithelial cell proliferation, inducing apoptosis,which plays a role in inhibiting the growth of ectopic lesions.
