Objective:1. The determination of MCP-1 expression quantity of glioma cell line (U87, U251 and T98G);2. The chemotaxis of U937 for glioma cell line (U87, U251 and T98G) in vitro;3. Through above two steps, provide experimental basis for further study the role of MCP-1 in glioma chemotaxis TAMs.Methods:1. By Western Blot method to measure the expression of MCP-1 for glioma cell line (U87,U251 and T98G);2. By Transwell method to observe the U937 cell chemotactic activity of glioma cell line (U87, U251 and T98G).Results:1. By Western Blot method to measure the MCP-1 grey value of glioma cell line (U87, U251 and T98G) and β-actin for the statistical analysis, the final result after comparing differences between each group had statistical significance (P< 0.05), and U87 group is higher than the U251, U251 above T98G group;2. By Transwell method observe the chemotaxis U937 cells of blank control and the glioma cell line (U87, U251 and T98G), on statistical analysis of difference between each group had statistical significance (P< 0.05), glioma cell line (U87, U251 and T98G) chemotaxis U937 cells were higher than blank control group, and U87 group is higher than the U251, U251 above T98G group.Conclusions:1. The glioma cell line (U87, U251 and T98G) all can express MCP-1, and the quantity of expression of MCP-1 differences between glioma cell lines;2. The glioma cell line (U87, U251 and T98G) can significantly chemotactic U937, and the quantity of chemotactic activity differences between glioma cell lines;3. The expression of MCP-1 and the chemotactic activity of glioma cell line (U87, U251 and T98G) are positive correlation, and provide experimental basis for further study the role of MCP-1 in glioma chemotaxis TAMs.
