Objective:To understand the expression of BKCa protein in human bladder smooth muscle cells (HBdSMC) and the effect of different extracellular concentrations 17-β estradiol on HBdSMC about the expression of BKCa protein and the cell current. Also to explore the correlation between 17-β estradiol, BKca and female OAB.Methods:We utilized the immunofluorescence technique to detect the differential expression of BKCa protein between HBdSMC cells with or without 17-β estradiol.Then detected the expression of BKCa protein in different concentraions of 17-β estradiol by western blotting.Next we used patch-clamp technique to detect the effect of 17-β estradiol on cell current.Results:The HBdSMC cells have the expression of BKCa protein,and 17-βestradiol can elevate its expression.And the level of BKCa protein has a positive correlation with extracellular estrogen concentration.17-β estradiol could increase the open probilitiy of BKca pathway,extend its open time,but not change the current amplitude.IBTX could inhibit the enhanced effect of 17-β estradiol on BKca pathway.Conclusions:HBdSMC cells can express BKCa protein.17-β estradiol can increase the expression of BKCa protein,enhance the cell BKca pathway current.This demonstrates that 17-β estradiol may promote the incidence of OAB in elderly female.
