ObjectiveCervical cancer is the most common malignancy of women in developing countries, the incidence of female genital malignant tumors accounted for the top. With the development of molecular biology in recent years, domestic and foreign research reports:Some types of potassium channels abundantly expressed in cervical cancer, and is closely related to the development of cervical cancer. Therefore, the potassium ion channel research has led to clinical attention. Potassium ion channels are embedded in the membrane lipids in the protein complex, application of molecular biology techniques to detect cell cycle proteins such as cyclinD, cell signaling pathway and DNA synthesis are all that potassium channel blockers can inhibit the progress of the cell cycle G1 phase.Calcium-activated potassium ion channels, currently known to exist three kinds of different sub-units:High-conductance calcium-activated potassium ion channels (BKCa), the conductance calcium-activated potassium ion channels (IKCa), low-conductance calcium-activated potassium ion channels (SKCa).Studies have reported that calcium-activated potassium ion channels in a variety of tumor cells to express, such as prostate cancer, glial cell tumors, stomach, pancreas, pituitary tumor, breast cancer, colorectal cancer and colon cancer.In the cervical cancer cell lines to express, and its expression in cervical cancer tissues and how, with cell proliferation, differentiation and the relationship between the prognosis is not clear.Therefore, this study selected BKCa was detected in normal cervical tissue and cervical cancer and cervical cancer radiotherapy and chemotherapy before and after the tissues, and explore the BKCa gene expression and cervical cancer occurrence and development of relations and their impact radiotherapy and chemotherapy.Materials and MethodsMaterial:Tissue used in this study were obtaines from 30 patients with invasive cervical cancer who underwent biopsy or curative resection, and another 10 pairs cervical cancer specimens from the same patients who before and after radioche motherapy at the First Affiliated Hospital of China Medical University. Besides, we collected 10 cases of normal cervical tissue. These tissue samples were obtained after hysterectomy because of benign uterine myoma. We collected the clinical information including pathological grades,clinical stages, age, before and after adiochemotherapy for statistical analysis.Main agents:RNAiso Reagent, TaKaRa RNA PCR Kit(AMV)Ver3.0, Forward and reverse primer of BKCa and P-actin, the necessary agents of agarose gel-electrophoresis.Method:RT-PCR Result judgement:BKCa mRNA amplified by PCR, agarose gelelectrophoresis,UV lamps observe results, and do semi-quantitative analysis by measuring the gray level.Statistical method:all data processed by SPSS13.0,measurement data was carried out by T-test.Differences were considered significant when P values were less than 0.05.Result1,The expression of BKCa mRNA in normal cervix and cervical cancerthe ratio of gray level of BKCa mRNA was2.2691 in cervical cancer,0.7322 in normal canvix.there were significant differences(P<0.01)2,Correlation between the expression of BKCa mRNA and clinicopathologic materials in patients with cervical cancerThe ratio of gray level of BKCa mRNA was 3.7866 in the poor differentiation group,higher than that of the middle and high differentiation group 2.6138,1.2165 (P<0.01). the ratio of gray level of BKCa mRNA was 1.0842 in stagesⅠ~Ⅱ,3.5688 in stagesⅢ-Ⅳ,there were significant differences(P<0.05). The expression of BKCa mRNA have no significant relation with age(P>0.05).3,The expression of BKCa mRNA in the same patient with cervical cancer before and after radio-chemotherapyBKCa mRNA was 2.3178 before radiotherapy and chemotherapy,0.4217 after radiotherapy and chemotherapy,they have significant differences(P<0.01).Conclusion1,BKCa mRNA participate in the proliferation of cervical cancer cell. 2,BKCa mRNA have a positive correlation with clinical stages and pathological grades, so they may be correlated with the malignant progression and prognosis of cervical cancer.3,The expression of BKCa mRNA in cervical cancer decrease after radiotherapy and chemotherapy.
