Objective Establishment of Sprague-Dawley rat model of subarachnoid hemorrha ge, by avoiding darkness test observation of subarachnoid hemorrhage model in rats at different time on learning and memory function, detection of synaptophysin expressio n in the model of subarachnoid hemorrhage in the hippocampus of rats, to explore th e relationship between P38and learning and memory.Methods1. Healthy male SD 120 rats were randomly divided into SAH model, SAH mod el group, sham operation group, normal control group, SAH model group were random 1y divided into 1d group,7d group,30 d group and 3 sub groups, model group inject ed from its tail artery blood, sham operation group to perform the same operation, injec tion of saline instead of bloodliquid,3 groups of rats fed with environment. The behavi ors of all rats were observed, with dark test method of recording every rat experiment allatency (latentperiod, LT) as well as the shuttle back and forth times (EN),study the a bility of learning and memory.2. The experimental group rats were made by single cage feeding, keeping the s ame environment, passive avoidance test after test, painless death SAH model group a nd 8 rats in the sham operation group and blank control group 8 rats, the remaining r ats into the next sub group. In SAH model group, every subgroup in 1d,7d,30 d was d ark after the experiment, rats were killed painless, model group every subgroup ratn=2 4, sham operation group and blank control group(n=24); detection of P38mRNA and pr otein expression. Each model of rats anesthetized with ether without the 2/3, RNA enzy me in hippocampus test stand out quickly,and liquid nitrogen frozen quickly moved to-80℃ refrigerator, another 1/3by inhalation of ether with 4% paraformaldehyde perfusio n, sucrose gradient after dehydration, the application of OTC embedded,-80℃ refrige rator for tissue sections. The relationship between the expression of RT-PCR by immu nohistochemistry, and Western blot technique in hippocampal P38mRNA of SAH mode lrats and related protein and to explore learning and memory.Result1. The success of the model was 97.3%. The rats in the blank group dark avoidance test results of LT was (53.00± 3.74s), EN (3.55 ± 1.51), model group, model group:after the success of the Id model. Through test of LT (37.86 ±3.57s), EN (5.71 ±0.49); SAH model group 7d dark avoidance test results LT (44.51±1.28s), EN (4.35± 1.19); SAH model group 30d avoid dark test results (51.73±3.62s), EN (3.89±1.07), sham operation group dark avoidance test results of LT was (53.14±3.11s), EN (3.55±1.51) times, the SAH model of rats in the model group compared with control group the learning and memory ability significantly decreased (P< 0.05, P< 0.01), the comparison between the two groups, the rats in the model group Id,7d, 30d group cognitive function increased, with statistical significance (P< 0.05, P< 0.01), but were higher than normal group difference the ratio, were statistically significant (P< 0.05, P< 0.01).2. molecular biology detection(1) immunohistochemical results:P38 immunohistochemical staining results showed hippocampal cells into brown. There were significant differences in model group and blank model and the percentage of positive cells of subarachnoid hemorrhage (P<0.05), model groupwas significantly lower than that of the control group. The model group,the percentage of positive cells of 1d was obviously lower than that of 7d group,30d group, 1d group was the lowest (P<0.05).(2) RT-PCR in the maker controls to strip is clearly visible, by measuring the optical density value of each band, and GADPH housekeeping gene ratio proofreading, there were significant differences in the model group and the blank of subarachnoid hemorrhage (P<0.05), the model group was significantly lower than that of the control group. Compared with model group, the optical density value of Id,7d proofreading lit group was significantly lower than that in 30d group, Id group was the lowest (P<0.05).(3) Western blot blotting results:the expression of P38 protein in hippocampus of rats in each group were compared in the standardprotein molecular, reference maker, through the determination of theobjective optical density with values, and P-actin housekeeping gene protein ratio proofreading, there were significant differences in the model group and the blank of subarachnoid hemorrhage (P<0.05), the model group was significantly lower than that of the control group. Compared with model group, the optical density value of Id,7d proofreading lit group was significantly higher in 30d group, 1d group was the lowest (P<0.05)Conclusion1.The cognitive function impaired learning memory of rats with subarachnoid hemorrhage.2. May be the molecular mechanism of impaired cognitive function changes of P38 expression in the hippocampus of rats with subarachnoid hemorrhage.
