Objective Stimulate Sprague-Dawley Rat Insular to establish chronic Epilepsy Model. To research the expression of activity-regulated cytoskeletal protein(Arc),brain-deried neurotrophic factor (BDNF), protein tyrosine kinases receptor B (TrkB)in Insula Chronic Electricity igniting epilepsy rat's Hippocampus, and to study the relation between Arc,BDNF,TrkB and electricity igniting epilepsy rat's learning and memory.Methods 1. To research relationship insular chronic electric igniting epilepsy rats and learning and memory : It need healthy adult male SD rats 216 ,to divide into model groups, sham operation groups, blank control groups, by building models of rat insula chronic electricity igniting, model groups divide into 0 week groups,1 week groups,2 week groups,3 week groups,4 week groups. to examine rats learning and memory ability by way of passive avoidance test. 2. Relationgship between insular epilepsy rat's hippocampus expression Arc,BDNF,TrkB and learning and memory :rats feeding, avoid single cage after passive avoidance test, no external stimuli painless kill the sham operation groups,and blank control groups rats. by way of passive avoidance test ,these rats is killed in different time in 1hour ,3 hour,6 hour ,12hour , 24hour .test the expression of Arc ,BDNF, TrkB in epilepsy both by immunohistochemistry S-P and western blot, and to test the expression of gene by RT-PCR. Result: There is difference in learning-remembering ability with different time for insula chronic electricity igniting rat. When igniting one week's time(W) , incubation period(LT:56.25±2.01 s) ,times of shuttling(EN: 3.42±0.93 times), the learning-remembering ability is no difference from that of blank comparison sets and false operation set, after 4 weeks(LT : 36.83±3.41 S , EN : 9.00±1.35 time) , there is obvious decline of learning-remembering(P<0.05). but when testing molecular biology of Arc, there is enhancement in expression of protein and gene for Arc in subgroup time from 1h., to the submit when 6h, and to primary level when 12h. The general tendency is to rise when one week(w), to rise fast when 2w-3w, to rise obviously slowly when 4w(P<0.05). As for the expression of BDNF and TrkB the peak value occurred in the 24 hours (P<0.05) within the experimental groups, it kept increasing in the 2nd and 3rd week and then dropped in the 4th week , but there was no statistical difference among the control and sham groups(P>0.05)Conclusion: Insular is an area that causes epilepsy symptom is proved by Chronic Epilepsy Model are succeeded by Stimulating Sprague-Dawley Rat Insular.Itself has the potential caused by epilepsy.The expression of Arc ,BDNF,TrkB rise first and decline afterwards in chronic insula electricity igniting rat Hippocampus, is parallel with the learning-remembering ability of epilepsy rats, Arc, BDNF,TrkB is the important factor of learning-remember ability for epilepsy rats.
