The incidence of middle ear tumor is low, however, the anatomy of the ear is very special, which is closed to brain tissue, so the middle ear tumor has more complications and high destruction. Otologist doctor need to pay more attention to ear tumor. Squamous carcinoma of the middle ear is one of malignant tumor occurred in middle ear. The early symptoms of squamous carcinoma of the middle ear are not obvious, which always accompanied by patients with the history of otitis media, so it need to be identifed with chronic suppurative otitis media. At present the cause of Squamous carcinoma of the middle ear is not clear, which may be associated to many factors. New blood vessels provide nutrients and oxygen to the tumor tissues, also provide the important way for tumor distant metastasis. It is closely related to the occurrence, development and prognosis of squamous carcinoma of the middle ear. Astrocytes increase gene-1(AEG-1) and Eph A7 are the two important genes involved in angiogenesis.AEG-1 is one of a newly discovered gene found in cancer recently,which is located in 8q22 chromosome and the length of c DNA is 3611 bp. The gene’s open reading is between 220 bp to 1968 bp, containing 12 exons and 11 introns and encoding an extremely conservative 80 k D protein. Eph receptor is divided into two major categories of Eph A and Eph B. Eph receptor and its Ephrin ligands control a series of interactions with different cells. Eph encode the protein which is a kind of the combination of membrane glycoprotein type I and contains the ligand binding area, functional areas and across membrane area of three parts and contains the structure area. Eph A7 encode the protein receptor tyrosine protein kinase which is one of the important members of this family.AEG-1 and Eph A7 are involved in the occurrence and development of tumor, regulation of intracellular signal transduction and transformation. Both of genes can participate in the formation of tumor blood vessels. However the expression and correlation of Eph A7 and AEG-1 have not been reported. Objective1. The expression of AEG-1 protein and Eph A7 protein in Squamous carcinoma of the middle ear and the normal ear skin tissue were detected.The different expression of AEG-1 protein and Eph A7 protein between Squamous carcinoma and the normal ear skin tissue were analyzed.2. To get acknowledge of pathogenetic mechanism of Squamous carcinoma of the middle ear, the expression of AEG-1 protein and Eph A7 protein in different pathological stage and clinical stage were detected. The role of Eph A7 and AEG-1 playing on the occurrence and the development process of the middle ear were analyzed.3. To investigate the correlation between AEG-1 protein and Eph A7 protein expressions, the inner mechanism was preliminary analyzed. of the middle ear had a positive correlation.The regulation of Eph A7 and AEG-1 may be inner linked.MaterialsThe paraffin-embeded samples were taken from the first affiliated hospital of zhengzhou university otolaryngology head and neck surgery surgical resection From January, 2007 to January, 2015(each samples were confirmed by two pathology experts),of which 21 smples were squamous cell carcinoma of the middle ear and 40 smples were normal ear skin tissue. MethodsThe paraffin-embeded samples were divided into group A and group B, stained with immunohistochemistry. Group A were squamous cell carcinoma of the middle ear, group B were normal ear skin tissue. Statistical dates were analyzed by SPSS17.0-statistical software. 1.The expression intensity of AEG-1 protein and Eph A7 protein in two groups were compared by using the method of t test. 2.The expression of AEG-1 protein and Eph A7 protein in different pathological stage and clinical stage were compared by using the c2 test. 3. The correlation of AEG-1 protein and Eph A7 protein were analyzed by using spearman-test. Results1. The Integral optical density(IOD) of AEG-1 protein in two groups were 125.17±22.25, 67.82±5.09, the IOD of group A compared to group B with a significant effect(P<0.05). The IOD of Eph A7 protein in two groups were 125.19±18.25, 65.82±7.09 with a significant effect(P<0.05).2. In Squamous carcinoma of the middle ear, the positive rate of AEG-1 protein in the pathological stage of T1-2 and T3-4 were 47.06%, 50.00%, which had no significant effect(p>0.05). The positive rate of Eph A7 protein in the pathological stage of T1-2 and T3-4 were 48.82%, 66.67%, which had no significant effect Materials(p>0.05).3. The positive rate of AEG-1 protein expression in Squamous carcinoma with lymph node metastasis was 87.50% and 38.46% without lymph metastasis.There is an evident difference between the two groups(p<0.05); The positive rate of Eph A7 protein expression in Squamous carcinoma of the middle ear with lymph node metastasis was 75.00% and 23.07% without lymph metastasis. There is an evident difference between the two groups(p<0.05).4. The positive rate of AEG-1 protein expression in the early and late clinical stage were 41.67% and 55.56%, and the two groups showed no significant difference(p>0.05). The positive rate of Eph A7 protein expressionin in the early and late clinical stage were 50.00%,44.44% with no significant difference(p> 0.05);5. There is a positive correlation between the expression of AEG-1 and Eph A7 protein in Squamous carcinoma of the middle ear(p< 0.05). Conclusions1. Our results showed the expression of AEG-1 protein and Eph A7 protein between group A compared to group B with a significant increasing. The expression of AEG-1 protein and Eph A7 protein in Squamous carcinoma with lymph node metastasis also increased. The expression of Eph A7 and AEG-1 may become one of the monitoring index of the middle ear cancer.2. The expression of AEG-1 protein and Eph A7 protein in the middle ear squamous carcinoma was accorded with pathological and clinical stage of tumor. AEG-1 protein and Eph A7 protein could play a positive role in promoting the occurrence,development and transfer of the middle ear squamous carcinomas.3.
