Allergy also named allergic reaction; it refers to a reaction by your immune system to antigen. Allergic reactions could lead to respiratory and cardiovascular disorder even can cause the shock. In recent years, the incidence of allergic reaction have been rising, it influenced the quality of people’s life, it also an important threats factor to human health. At present, there also lack of a comprehensive diagnosis and treatment system to allergic reaction, clinical diagnosis is usually based on the patient’s history, which is easy lead to false positive. So far, there is none kind of medicine can completely cure allergic disease without no side effects and no recrudescence. Therefore, it has extremely great significance to looking for a new accurate allergy diagnosis method and safety natural allergy treatment.Metabonomics as a new discipline has developed in recent years. It mainly analyzes the change of small molecule metabolite in the body by using modern high precision analysis instrument, which was influenced by environmental, chemical. The metabolism matains a dynamic balance in body, any disease would break its balance resulting in obvious changes of metabolites. Therefore, metabonomics is a method which could be able to find the biomarkers of disease, which will be helpful to the diagnosis, prevention, treatment and prognosis judgement.In this research, a SD rat allergic model was built by injecting ovalbumin. Allergic biomarkers were found in this study. The anti-allergic effect o ganoderma lucidum (3-glucan, yeast β-glucan and Resveratrol were analyzed by using compared the metabolites of different groups in urine and serum, the anti-allergy mechanism were also discussed in this paper. Study results were show as follows:1. Allergy model buildThe allergy effect of different ovalbumin concentrations were compared by symptoms and IgE concentration in serum. The allergy model was built by using20mg/mL ovalbumin.2. Q-Tof detection methodFor the sample pretreatment method, we compared methanol, acetonitrile and the mixture of methanol and acetonitrile. Acetonitrile was determined to be used as a deproteinization reagent. Three times was better than others when compared different dilution ratio.As to the buffer in mobile phase, three different buffer solutions were compared. It showed that ammonium formate may inhibit the ionization of samples. Acetic acid is relatively well compared to formic acid, and especially the0.2%acetic acid which was chosen as a composition in mobile phase. Finally, flow rate was set0.35mL/min and the analysis time was35min after optimization.3. The metabolomics research of allergyWhen used as anti-allergy substances, it suggested the anti-allergic effect of resveratrol is better, followed by yeast β--glucan and ganoderma lucidum β-glucan. The anti-allergy was mainly through regulating immune cells and free radical scavenging.
